Description
Overview of the medicine
Tamsulosin is an alpha-blocker used to treat symptoms of an enlarged prostate (benign prostatic hyperplasia or BPH) such as difficulty urinating, weak stream, and frequent urination. It works by relaxing muscles in the prostate and bladder neck.
Uses & Indications
Dosage
Adults
0.4 mg once daily, taken approximately 30 minutes after the same meal each day. May be increased to 0.8 mg once daily after 2-4 weeks if necessary.
Elderly
No dosage adjustment is generally required.
Renal_impairment
No dosage adjustment is generally required for mild to moderate impairment; use with caution in severe impairment.
How to Take
Take orally, once daily, approximately 30 minutes after the same meal each day. Do not crush, chew, or open the tablet.
Mechanism of Action
Tamsulosin selectively and competitively binds to alpha-1A adrenergic receptors in the prostate, bladder neck, and prostatic urethra, causing relaxation of the smooth muscle in these tissues. This decreases outflow obstruction and improves urine flow.
Pharmacokinetics
Onset
Symptomatic improvement within 1-2 weeks, full effect may take longer.
Excretion
Primarily via urine (76%) and feces (21%) as metabolites and unchanged drug.
Half life
Approximately 9-15 hours.
Absorption
Well absorbed from the gastrointestinal tract, nearly 90% bioavailable. Food affects absorption; typically taken 30 minutes after the same meal each day.
Metabolism
Primarily hepatic, extensive metabolism by CYP3A4 and CYP2D6.
