Description
Overview of the medicine
Vaxitet 40 IU Injection is a preparation of human tetanus immunoglobulin (TIG) for intramuscular administration. It provides immediate, passive immunity against tetanus for individuals at risk, particularly in specific low-risk scenarios or as a component of broader prophylaxis measures.
Uses & Indications
Dosage
Adults
The standard prophylactic dose for tetanus immunoglobulin (TIG) is typically 250 IU to 500 IU. Vaxitet 40 IU is intended for specific, very low-risk prophylactic scenarios or as determined by local clinical guidelines where a lower dose is deemed sufficient. Administer 40 IU (1 vial) intramuscularly, as a single dose.
Elderly
No specific dose adjustment required for elderly patients; dose as per adult guidelines for specific indications.
Renal_impairment
No specific dose adjustment required for renal impairment, as immunoglobulins are naturally processed.
How to Take
Vaxitet 40 IU Injection is for intramuscular administration only. Inject slowly into a large muscle mass, such as the deltoid or gluteus maximus. Do not administer intravenously.
Mechanism of Action
Tetanus immunoglobulin contains antibodies against tetanus toxin. These antibodies neutralize the tetanus toxin produced by Clostridium tetani, thereby preventing the toxin from binding to nerve endings and causing neurological symptoms associated with tetanus.
Pharmacokinetics
Onset
Passive immunity is immediate, with measurable antibody levels within hours.
Excretion
Degraded into small peptides and amino acids, then excreted.
Half life
The half-life of human IgG in individuals with normal IgG catabolism is approximately 3-4 weeks (21-28 days).
Absorption
Tetanus immunoglobulin is administered intramuscularly and is slowly absorbed into the circulation. Peak serum levels are usually achieved within 2-3 days.
Metabolism
Immunoglobulins are catabolized in the same way as endogenous IgG molecules, primarily in reticuloendothelial cells.
Side Effects
Contraindications
- •Known hypersensitivity to human immunoglobulin or any component of the preparation.
- •Individuals with selective IgA deficiency who have known antibodies against IgA, as severe anaphylactic reactions may occur.
Drug Interactions
Live attenuated vaccines (e.g., Measles, Mumps, Rubella, Varicella)
Administration of immunoglobulin may interfere with the immune response to live attenuated vaccines. It is generally recommended to wait at least 3 months after TIG administration before giving live attenuated vaccines. Consult vaccine guidelines.
Storage
Store at 2°C to 8°C (refrigerate). Do not freeze. Protect from light. Do not use if the solution is turbid or contains particles.
Overdose
No specific symptoms of overdose have been reported. Even if a higher dose than recommended is accidentally administered, it is unlikely to cause adverse effects beyond those associated with the administration of large protein volumes (e.g., local discomfort).
Pregnancy & Lactation
Pregnancy Category C. Tetanus immunoglobulin has been widely used for decades in pregnant and lactating women, and there is no evidence of adverse effects on the course of pregnancy, the fetus, or the neonate. It is considered safe when clinically indicated.
