Overview of the medicine
Veridipin-Plus is a combination medicine containing Amlodipine, a calcium channel blocker, and Valsartan, an angiotensin II receptor blocker. It is used to treat high blood pressure (hypertension) in patients whose blood pressure is not adequately controlled with either component alone.
Uses & Indications
Dosage
Adults
Usually one Veridipin-Plus 5 mg tablet (Amlodipine 5 mg + Valsartan 80 mg) once daily. Dosage should be individualized based on response and tolerability.
Elderly
No specific dosage adjustment is required, but caution is advised. Initiate with the lowest effective dose.
Renal_impairment
No initial dosage adjustment is needed for mild to moderate renal impairment. Caution is advised in severe renal impairment.
How to Take
Take the tablet orally once daily, with or without food. Swallow the tablet whole with water. It is recommended to take it at approximately the same time each day.
Mechanism of Action
Amlodipine works by relaxing blood vessels, making it easier for the heart to pump blood. Valsartan blocks the action of a natural substance in the body (angiotensin II) that constricts blood vessels, thereby lowering blood pressure. The combination provides additive blood pressure lowering effects.

Amlodipine: Primarily renal (inactive metabolites). Valsartan: Primarily fecal (unmetabolized drug).

Amlodipine: Well absorbed (bioavailability 64-90%). Valsartan: Rapidly absorbed (bioavailability ~23%).
Metabolism
Amlodipine: Extensively metabolized in the liver to inactive metabolites. Valsartan: Minimally metabolized (only 20% by the liver).
Side Effects
Contraindications
- •Hypersensitivity to amlodipine, valsartan, or any component of the formulation.
- •Pregnancy (second and third trimesters) due to fetal toxicity.
- •Severe hepatic impairment or biliary cirrhosis.
- •Concomitant use with aliskiren in patients with diabetes or renal impairment (GFR < 60 mL/min/1.73 m²).
Drug Interactions
Lithium
Increased serum lithium concentrations and toxicity have been reported with ARBs.
NSAIDs (Nonsteroidal Anti-inflammatory Drugs)
May reduce the antihypertensive effect and increase the risk of renal impairment.
Potassium-sparing diuretics/Potassium supplements
May lead to hyperkalemia (increased potassium levels).
CYP3A4 inducers (e.g., rifampicin, St. John's Wort)
May decrease amlodipine exposure, reducing efficacy.
CYP3A4 inhibitors (e.g., ketoconazole, clarithromycin)
May increase amlodipine exposure, leading to increased risk of side effects.
Storage
Store below 30°C in a dry place, away from direct light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include marked hypotension (low blood pressure) and reflex tachycardia. Management involves symptomatic and supportive treatment. If recent ingestion, gastric lavage may be considered. Intravenous fluids and vasopressors may be needed for hypotension.
Pregnancy & Lactation
Pregnancy: Contraindicated during the second and third trimesters due to potential fetal harm. Discontinue immediately if pregnancy is detected. Lactation: Not recommended. It is unknown if amlodipine or valsartan is excreted in human breast milk.
