Description
Overview of the medicine
Verivas 5 mg Tablet contains Solifenacin Succinate, which is used to treat symptoms of overactive bladder, such as urinary urgency, frequency, and urge incontinence. It works by relaxing the bladder muscles, helping to control urination.
Uses & Indications
Dosage
Adults
The recommended starting dose is 5 mg once daily. If needed, the dose may be increased to 10 mg once daily.
Elderly
No dosage adjustment is required in elderly patients.
Renal_impairment
For patients with severe renal impairment (creatinine clearance < 30 mL/min), a dose greater than 5 mg is not recommended.
How to Take
Take the tablet orally once daily with water, with or without food. Swallow the tablet whole; do not crush, chew, or divide it.
Mechanism of Action
Solifenacin is a competitive muscarinic receptor antagonist. It inhibits muscarinic M3 receptors in the bladder, which are primarily responsible for bladder smooth muscle contraction. By blocking these receptors, it reduces bladder muscle activity, thereby increasing bladder capacity and decreasing the frequency of urination, urgency, and incontinence episodes.
Pharmacokinetics
Onset
Clinical effects usually observed within 1 week, with full effect within 4 weeks.
Excretion
Eliminated mainly in the urine (about 70%) and feces (about 15-20%).
Half life
Approximately 45-68 hours.
Absorption
Well absorbed from the gastrointestinal tract, reaching peak plasma concentrations in 3-8 hours. Bioavailability is about 90%.
Metabolism
Extensively metabolized in the liver, primarily by cytochrome P450 3A4 (CYP3A4) enzyme. Active metabolites exist but contribute minimally to clinical effect.

Drug Interactions
Anticholinergic agents
Concomitant use with other anticholinergic agents may lead to more pronounced anticholinergic adverse effects.
Cholinergic receptor agonists
May reduce the effect of cholinergic receptor agonists.
CYP3A4 Inhibitors (e.g., Ketoconazole)
Co-administration with strong CYP3A4 inhibitors like ketoconazole may increase solifenacin exposure, requiring a dose reduction of solifenacin to 5 mg.

Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include severe anticholinergic effects such as dry mouth, blurred vision, constipation, urinary retention, and tachycardia. Treatment should be symptomatic and supportive. Gastric lavage and activated charcoal may be considered. Physostigmine can be used for severe anticholinergic symptoms.
Pregnancy & Lactation
Pregnancy Category C. Use only if the potential benefit justifies the potential risk to the fetus. It is not known if solifenacin is excreted in human milk; therefore, caution should be exercised when administered to a nursing mother.
