Overview of the medicine
Vonolend 20 mg Tablet contains Vonoprazan, a potassium-competitive acid blocker (P-CAB) used to treat acid-related disorders such as gastric ulcers, duodenal ulcers, and reflux esophagitis, and as an adjunct in H. pylori eradication therapy.
Uses & Indications
Dosage
Adults
Gastric ulcer: 20 mg once daily for 8 weeks. Duodenal ulcer: 20 mg once daily for 6 weeks. Reflux esophagitis: 20 mg once daily for 4 weeks (up to 8 weeks if necessary). H. pylori eradication: 20 mg twice daily in combination with two antibiotics for 7 days.
Elderly
No specific dosage adjustment is usually required, but monitor for adverse effects.
Renal_impairment
No dosage adjustment needed for mild to moderate renal impairment. Use with caution and monitor closely in severe renal impairment.
How to Take
Take orally, with or without food. For H. pylori eradication, take with food. Usually taken once daily, preferably in the morning before breakfast.
Mechanism of Action
Vonoprazan acts by reversibly inhibiting the H+/K+-ATPase enzyme (proton pump) in the gastric parietal cells, thereby blocking the final step of acid secretion into the stomach lumen. Unlike traditional PPIs, it does not require acid activation and binds to the pump regardless of its activation state.

Onset of acid suppression occurs within hours of the first dose.

Approximately 50% excreted via urine and 50% via feces.
Half life
Plasma elimination half-life is approximately 7-9 hours.
