Description
Overview of the medicine
Amifen 50 mg Suspension contains Mefenamic Acid, a non-steroidal anti-inflammatory drug (NSAID) used to relieve mild to moderate pain, inflammation, and fever, especially in children. It is also used for symptomatic relief of primary dysmenorrhea and menorrhagia.
Uses & Indications
Dosage
Adults
Not typically used for adults in this suspension form. For adults, usually 500 mg initially, then 250 mg every 6 hours as needed.
Elderly
Use with caution; lower doses may be required due to increased risk of adverse effects.
Renal_impairment
Contraindicated in severe renal impairment. Use with caution in mild to moderate impairment, dose adjustment may be necessary.
How to Take
Take orally, preferably with or after food, to minimize gastrointestinal discomfort. Shake well before use. Use a measuring spoon or cup for accurate dosing.
Mechanism of Action
Mefenamic Acid inhibits cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2) enzymes, which are responsible for the synthesis of prostaglandins. Prostaglandins mediate pain, inflammation, and fever, so by inhibiting their synthesis, Mefenamic Acid reduces these symptoms.

Onset
Within 30-60 minutes for analgesic effects.
Excretion
Mainly excreted in urine (52-67%) and feces (20-25%), primarily as metabolites.
Half life
Approximately 2-4 hours.
Absorption
Readily and well absorbed from the gastrointestinal tract. Peak plasma concentrations are achieved in 2-4 hours.
Metabolism
Extensively metabolized in the liver by cytochrome P450 enzyme CYP2C9 to 3-hydroxymethyl mefenamic acid and 3-carboxy mefenamic acid.
Side Effects
Contraindications
- •Hypersensitivity to Mefenamic Acid, aspirin, or other NSAIDs
- •Active gastrointestinal ulceration, inflammation, or bleeding
- •Severe renal or hepatic impairment
- •Severe heart failure
- •Patients with a history of aspirin-induced asthma, urticaria, or allergic-type reactions
- •Third trimester of pregnancy
Drug Interactions
Lithium
Increased plasma lithium levels, leading to toxicity.
Methotrexate
Increased methotrexate levels, leading to toxicity.
Corticosteroids
Increased risk of gastrointestinal ulceration and bleeding.
Diuretics (e.g., Furosemide)
Reduced diuretic and antihypertensive effects, increased risk of renal impairment.
Anticoagulants (e.g., Warfarin)
Increased risk of bleeding due to inhibition of platelet aggregation and gastric ulceration.
ACE inhibitors/Angiotensin II receptor blockers
Reduced antihypertensive effect and increased risk of renal impairment.
Storage
Store below 30°C in a dry place, away from light and moisture. Do not freeze. Keep out of reach of children.
Overdose
Symptoms of overdose may include lethargy, drowsiness, nausea, vomiting, epigastric pain, and gastrointestinal bleeding. Rarely, hypertension, acute renal failure, respiratory depression, and coma may occur. Management: Gastric lavage, activated charcoal, and symptomatic and supportive treatment. There is no specific antidote.
Pregnancy & Lactation
Pregnancy: Contraindicated in the third trimester due to potential adverse effects on the fetal cardiovascular system (premature closure of ductus arteriosus). Use in early pregnancy only if clearly needed and benefit outweighs risk. Lactation: Mefenamic Acid is excreted in breast milk in small amounts. Use with caution during breastfeeding; consult a doctor.
