Description
Overview of the medicine
Amoxicillin is a penicillin antibiotic used to treat a variety of bacterial infections.
Uses & Indications
Dosage
Adults
250 mg to 500 mg every 8 hours, or 500 mg to 875 mg every 12 hours, depending on infection severity.
Elderly
Similar to adult dosage, but caution with renal impairment.
Renal_impairment
Dosage adjustment required based on creatinine clearance (e.g., lower dose or extended interval).
How to Take
Oral, can be taken with or without food. Complete the full course of treatment.
Mechanism of Action
Inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs).
Pharmacokinetics
Onset
Rapid, typically within 1-2 hours.
Excretion
Primarily via renal excretion (glomerular filtration and tubular secretion).
Half life
Approximately 1-1.5 hours in adults with normal renal function.
Absorption
Well absorbed from the gastrointestinal tract, not significantly affected by food.
Metabolism
Minimally metabolized; about 10-25% is hydrolyzed to an inactive penicilloic acid.
Side Effects
Contraindications
- •Hypersensitivity to amoxicillin or other penicillins
- •Severe allergic reactions to other beta-lactam antibiotics
Drug Interactions
Probenecid
Increases and prolongs amoxicillin blood levels by reducing renal tubular secretion.
Methotrexate
Increased methotrexate toxicity due to reduced renal clearance.
Oral contraceptives
May reduce efficacy of oral contraceptives (rarely).
Oral anticoagulants (e.g., Warfarin)
May prolong prothrombin time; monitor INR.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms may include gastrointestinal upset, rash, or convulsions in severe cases. Management is supportive; hemodialysis can remove amoxicillin from circulation.
Pregnancy & Lactation
Generally considered safe during pregnancy (Category B). Excreted in breast milk in small amounts; generally considered safe for nursing infants, but monitor for diarrhea or candidiasis.
