Overview of the medicine
Arotrix (Etoricoxib) is a selective cyclooxygenase-2 (COX-2) inhibitor NSAID used to relieve pain and inflammation in conditions like osteoarthritis, rheumatoid arthritis, ankylosing spondylitis, and acute gouty arthritis.
Uses & Indications
Dosage
Adults
For Osteoarthritis: 60 mg once daily. For Rheumatoid arthritis and Ankylosing spondylitis: 90 mg once daily. For Acute gouty arthritis: 120 mg once daily (maximum 8 days). For Acute pain and Primary dysmenorrhea: 120 mg once daily (maximum 8 days).
Elderly
No dose adjustment is generally required in elderly patients, but caution should be exercised.
Renal_impairment
No dose adjustment needed for mild to moderate renal impairment. Not recommended for severe renal impairment.
How to Take
Arotrix tablets can be taken orally with or without food.
Mechanism of Action
Etoricoxib selectively inhibits the cyclooxygenase-2 (COX-2) enzyme, which is responsible for the synthesis of prostaglandins, mediators of pain and inflammation. By inhibiting COX-2, it reduces prostaglandin production, thereby alleviating pain and inflammation.

Onset
Onset of action typically within 24 minutes.
Excretion
Approximately 70% of the dose is excreted in the urine and 20% in the feces, predominantly as metabolites.
Half life
The elimination half-life is approximately 22 hours.
Absorption
Rapidly and well absorbed after oral administration, with high absolute bioavailability (approximately 100%).
Metabolism
Extensively metabolized in the liver, primarily via cytochrome P450 (CYP3A4) to inactive metabolites.
Side Effects
Contraindications
- •Hypersensitivity to etoricoxib or any component of the formulation
- •Active peptic ulceration or gastrointestinal bleeding
- •Severe hepatic dysfunction
- •Severe renal impairment
- •Congestive heart failure (NYHA II-IV)
- •Established ischemic heart disease, peripheral arterial disease, and/or cerebrovascular disease
- •Uncontrolled hypertension
Drug Interactions
Lithium
May increase plasma lithium levels, leading to toxicity.
Warfarin
May increase the risk of bleeding.
ACE inhibitors/Angiotensin II antagonists
May reduce the antihypertensive effect and increase the risk of renal impairment.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include gastrointestinal disturbances (e.g., nausea, vomiting) and renal toxicity. Management is supportive and symptomatic.
Pregnancy & Lactation
Arotrix is not recommended during pregnancy (Pregnancy Category C) or lactation due to potential risks to the fetus and infant.
