Description
Overview of the medicine
Artigo is a combination medication containing Cinnarizine and Dimenhydrinate, primarily used to treat symptoms of vertigo, dizziness, nausea, and vomiting associated with various vestibular disorders, including Meniere's disease.
Uses & Indications
Dosage
Adults
One tablet (Cinnarizine 20 mg + Dimenhydrinate 40 mg) three times daily after meals. The maximum daily dose should not exceed 3 tablets.
Elderly
Same as adults, but caution is advised due to increased sensitivity to anticholinergic effects and sedation. Dose adjustment might be considered if necessary.
Renal_impairment
Use with caution. Dose adjustment may be required in severe renal impairment. Consult a physician.
How to Take
Oral administration. Tablets should be taken after meals with water to reduce gastrointestinal irritation.
Mechanism of Action
Cinnarizine, a calcium channel blocker and antihistamine, reduces vestibular excitability. Dimenhydrinate, an antihistamine with anticholinergic properties, provides antiemetic and sedative effects, acting on the chemoreceptor trigger zone and vestibular apparatus.
Pharmacokinetics
Onset
Onset of action typically occurs within 30-60 minutes.
Excretion
Metabolites are primarily excreted via urine and feces.
Half life
Cinnarizine has a half-life of approximately 4 hours, and dimenhydrinate has a half-life of 4-6 hours.
Absorption
Both cinnarizine and dimenhydrinate are well absorbed from the gastrointestinal tract. Peak plasma levels are reached within 1-4 hours.
Metabolism
Both components are extensively metabolized in the liver.
Side Effects
Contraindications
- Hypersensitivity to cinnarizine, dimenhydrinate, or other antihistamines
- Acute porphyria
- Angle-closure glaucoma
- Prostatic hypertrophy
- Epilepsy
- Parkinson's disease (due to potential for exacerbation of symptoms)
Drug Interactions
Ototoxic drugs (aminoglycosides)
Artigo may mask the warning signs of ototoxicity, such as dizziness or tinnitus.
CNS depressants (alcohol, sedatives, hypnotics)
Potentiated sedative effects, increased risk of CNS depression.
Anticholinergic drugs (tricyclic antidepressants, MAO inhibitors)
Increased anticholinergic effects like dry mouth, blurred vision, urinary retention.
Storage
Store in a cool, dry place below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include drowsiness, dizziness, ataxia, dry mouth, blurred vision, tachycardia, urinary retention, and in severe cases, seizures, coma, and respiratory depression. Treatment is symptomatic and supportive, including gastric lavage and activated charcoal.
Pregnancy & Lactation
Not recommended during pregnancy due to insufficient safety data. Dimenhydrinate is excreted in breast milk; therefore, use is not recommended during lactation.
