Description
Overview of the medicine
Asitrol 10 mg Tablet contains Acidoxime, a proton pump inhibitor (PPI) used to reduce stomach acid production. It is prescribed for conditions like gastroesophageal reflux disease (GERD), peptic ulcers, and Zollinger-Ellison syndrome.
Uses & Indications
Dosage
Adults
10-20 mg once daily, preferably before a meal. For GERD, 10 mg once daily for 4-8 weeks. For PUD, 20 mg once daily for 4 weeks.
Elderly
No specific dose adjustment required, but caution is advised due to potential for age-related decline in renal/hepatic function.
Renal_impairment
No dose adjustment is generally required for mild to moderate renal impairment. For severe impairment, caution is advised.
How to Take
Take the tablet whole with water, preferably in the morning before breakfast. Do not crush, chew, or break the tablet.
Mechanism of Action
Acidoxime selectively inhibits the H+/K+-ATPase enzyme system (the proton pump) at the secretory surface of the gastric parietal cells. This action blocks the final step of acid production, thereby reducing gastric acidity.
Pharmacokinetics
Onset
Within 1-2 hours, maximal effect within 3-5 days of continuous use.
Excretion
Mainly through urine (approx. 80%) and feces (approx. 20%).
Half life
Approximately 1-1.5 hours.
Absorption
Rapidly absorbed after oral administration; bioavailability is approximately 50-60%.
Metabolism
Extensively metabolized in the liver, primarily by CYP2C19 and CYP3A4 enzymes.
Side Effects
Contraindications
- •Hypersensitivity to Acidoxime or any component of the formulation
- •Concomitant use with nelfinavir
Drug Interactions
Warfarin
Increased INR/prothrombin time; monitor closely.
Clopidogrel
May reduce the antiplatelet effect of clopidogrel by inhibiting CYP2C19.
Methotrexate
Increased plasma levels of methotrexate, especially at high doses.
Storage
Store below 30°C in a dry place, away from direct light and moisture. Keep out of reach of children.
Overdose
Symptoms are generally mild and include nausea, vomiting, dizziness, abdominal pain, diarrhea, and headache. No specific antidote; treatment is symptomatic and supportive.
Pregnancy & Lactation
Use during pregnancy only if clearly needed and the potential benefits outweigh the risks. Excretion into breast milk is unknown; caution advised in nursing mothers.
