Overview of the medicine
Avidro 05 mg Tablet contains Avidralactone, an angiotensin II receptor blocker (ARB) used to treat high blood pressure (hypertension) and heart failure. It helps to relax blood vessels, allowing blood to flow more easily and thereby lowering blood pressure.
Uses & Indications
Dosage
Adults
Initial dose is usually 5 mg once daily. May be increased up to 40 mg daily depending on blood pressure response and tolerability. Maximum dose 80 mg daily.
Elderly
No specific dose adjustment is usually required, but caution is advised in very elderly patients due to potential increased sensitivity.
Renal_impairment
No initial dose adjustment is needed for mild to moderate renal impairment. For severe renal impairment, consider a lower starting dose (e.g., 2.5 mg) and careful monitoring.
How to Take
Administer orally, once daily, with or without food. Swallow the tablet whole with water.
Mechanism of Action
Avidralactone selectively blocks the binding of angiotensin II to the AT1 receptor, which is found in many tissues, including vascular smooth muscle and the adrenal gland. This inhibition prevents the vasoconstrictive and aldosterone-secreting effects of angiotensin II, leading to vasodilation and reduced blood pressure.

Onset
Antihypertensive effect usually starts within 1-2 hours, with full effect seen within 2-4 weeks of continuous therapy.
Excretion
Mainly excreted unchanged in urine and feces.

Approximately 6-9 hours.

Rapidly absorbed after oral administration, with peak plasma concentrations reached within 1-2 hours. Bioavailability is approximately 30-40%.

Side Effects
Contraindications
- Hypersensitivity to Avidralactone or any component of the formulation
- Pregnancy (especially second and third trimesters)
- Concomitant use with aliskiren in patients with diabetes or renal impairment (GFR < 60 mL/min/1.73 m²)
Drug Interactions
Lithium
May increase serum lithium concentrations and lead to lithium toxicity.
NSAIDs (e.g., ibuprofen, naproxen)
May reduce the antihypertensive effect and worsen renal function in dehydrated or elderly patients.
Potassium-sparing diuretics or potassium supplements
May lead to increased serum potassium levels (hyperkalemia).
Storage
Store in a cool, dry place below 30°C, away from direct sunlight and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include profound hypotension (low blood pressure) and tachycardia; bradycardia could occur from parasympathetic stimulation. Treatment is symptomatic and supportive. Administer activated charcoal shortly after ingestion. Intravenous normal saline infusion may be required to support blood pressure.
Pregnancy & Lactation
Category D in pregnancy (second and third trimesters) due to fetal toxicity. Not recommended during breastfeeding as it is unknown if Avidralactone is excreted in human milk. Consult a doctor immediately if you are pregnant or planning to breastfeed.
