Description
Overview of the medicine
Azobrin (Brinzolamide) is a topical carbonic anhydrase inhibitor used to lower elevated intraocular pressure in patients with ocular hypertension or open-angle glaucoma.
Uses & Indications
Dosage
Adults
One drop in the affected eye(s) three times daily.
Elderly
No dosage adjustment required for elderly patients.
Renal_impairment
Use with caution in patients with severe renal impairment (creatinine clearance <30 mL/min). Not recommended in end-stage renal disease.
How to Take
For ophthalmic use only. Shake well before use. Apply one drop into the conjunctival sac of the affected eye(s). Avoid touching the dropper tip to the eye or any other surface.
Mechanism of Action
Brinzolamide inhibits carbonic anhydrase in the ciliary body of the eye. This inhibition reduces the formation of bicarbonate ions, which in turn reduces sodium and fluid transport, thereby decreasing aqueous humor secretion and lowering intraocular pressure.

Onset
Onset of IOP lowering effect is typically within 2 hours.
Excretion
Excreted predominantly by the kidneys, primarily as unchanged drug and its metabolite.
Half life
The apparent half-life in red blood cells is approximately 111 days.
Absorption
After ocular administration, brinzolamide is absorbed into the systemic circulation. It binds extensively to carbonic anhydrase in red blood cells.
Metabolism
Metabolized primarily to N-desethylbrinzolamide, which also binds to carbonic anhydrase.
Side Effects
Contraindications
- Hypersensitivity to the active substance or to any of the excipients
- Hypersensitivity to sulfonamides
- Severe renal impairment (creatinine clearance <30 mL/min)
- Hyperchloraemic acidosis

Pregnancy & Lactation
Pregnancy Category C. Should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. It is not known whether brinzolamide is excreted in human milk; caution should be exercised when administered to a nursing woman.
