Description
Overview of the medicine
Azole 200 mg Suspension is an antifungal medication used to treat a variety of fungal infections, including systemic candidiasis, cryptococcosis, and fungal infections of the skin, nails, and mucous membranes. It works by inhibiting the growth of fungi.
Uses & Indications
Dosage
Adults
For most systemic candidiasis, 200 mg on day 1, then 100-200 mg daily for 2-4 weeks. For cryptococcal meningitis, 400 mg on day 1, then 200-400 mg daily for 6-8 weeks.
Elderly
No dosage adjustment needed unless renal function is impaired.
Renal_impairment
Dosage should be reduced based on creatinine clearance (e.g., 50% reduction for CrCl < 50 mL/min).
How to Take
Shake the bottle well before each use. Administer orally, with or without food. Use a calibrated measuring device for accurate dosing.
Mechanism of Action
Azole inhibits cytochrome P450-dependent 14-alpha-demethylase, an enzyme crucial for fungal ergosterol synthesis. This disruption of ergosterol, a vital component of the fungal cell membrane, leads to increased permeability and eventual cell death.
Pharmacokinetics
Onset
Antifungal effects typically seen within several days of initiation, full effect may take longer.
Excretion
Primarily excreted unchanged in urine (80% of dose).
Half life
Approximately 30 hours, allowing for once-daily dosing.
Absorption
Well absorbed orally, bioavailability is >90%. Peak plasma concentrations reached within 1-2 hours.
Metabolism
Minimal hepatic metabolism (approx. 11% of dose), primarily by CYP2C9 and CYP2C19.
