Description
Overview of the medicine
Baclofen is a centrally acting skeletal muscle relaxant used to treat spasticity resulting from multiple sclerosis, spinal cord injuries, and other spinal cord diseases.
Uses & Indications
Dosage
Adults
Initial dose: 5 mg three times daily. May be increased by 5 mg every 3 days to a maximum of 80 mg/day (20 mg four times daily).
Elderly
Start with lower doses and titrate slowly due to increased sensitivity and potential for adverse effects.
Renal_impairment
Dose should be reduced in patients with renal impairment, with careful monitoring.
How to Take
Administer orally, preferably with food or milk to minimize gastric irritation.
Mechanism of Action
Baclofen acts as a GABA-B receptor agonist, leading to hyperpolarization of neurons and a reduction in excitatory neurotransmitter release, thereby decreasing muscle spasticity.

Contraindications

Drug Interactions
Tricyclic Antidepressants (TCAs)
May potentiate muscle relaxant effects and cause pronounced hypotonia.
CNS Depressants (e.g., alcohol, opioids, benzodiazepines)
Increased CNS depression, drowsiness, and respiratory depression.
Storage
Store at controlled room temperature (20-25°C), away from moisture and direct light. Keep out of reach of children.
Overdose
Symptoms include drowsiness, coma, respiratory depression, seizures, and hypothermia. Management involves supportive care, gastric lavage if recent ingestion, and respiratory support as needed.
