Overview of the medicine
Baclofen 5 mg Tablet is a muscle relaxant used to treat muscle spasms, stiffness, and pain caused by certain conditions like multiple sclerosis, spinal cord injury, or other neurological disorders. It works by calming overactive nerves in the brain and spinal cord that control muscle movement.
Uses & Indications
Dosage
Adults
Initial dose is 5 mg three times daily (TID) for 3 days. Dosage may be increased by 5 mg TID increments every 3 days until optimal response is achieved. Maximum recommended daily dose is 80 mg (e.g., 20 mg four times daily).
Elderly
Start with a low dose (e.g., 5 mg once or twice daily) and increase gradually to minimize adverse effects, as elderly patients may be more sensitive to the effects.
Renal_impairment
Lower doses are required in patients with renal impairment. Dosage should be reduced significantly, and caution is advised. Consult a doctor for specific dosing.
How to Take
Take Baclon 5 mg Tablet orally, usually with food or milk to reduce gastrointestinal upset. Swallow the tablet whole with water. Do not crush, chew, or break it.
Mechanism of Action
Baclofen is a gamma-aminobutyric acid (GABA) derivative that acts as a GABA-B receptor agonist. It inhibits both mono- and polysynaptic reflexes at the spinal level, leading to reduced spasticity by hyperpolarizing afferent terminals and inhibiting the release of excitatory neurotransmitters.

Onset
Onset of action is usually within 0.5 to 1 hour.
Excretion
Primarily excreted unchanged by the kidneys (70-80% within 72 hours). A small amount is excreted in feces.
Half life
Plasma elimination half-life is approximately 2.5 to 4 hours.
Absorption
Rapid and extensive absorption from the gastrointestinal tract. Peak plasma concentrations are achieved within 2-3 hours.
Metabolism
Minimal hepatic metabolism; about 15% is metabolized by deamination.
Side Effects
Contraindications

Drug Interactions
Antihypertensives
May cause an additive hypotensive effect, leading to lower blood pressure.
Levodopa/Carbidopa
Possible increased risk of confusion, hallucinations, and agitation.
Tricyclic Antidepressants (TCAs)
May potentiate the effects of Baclofen, leading to increased muscle hypotonia.
Monoamine Oxidase Inhibitors (MAOIs)
Potential for increased CNS depression and hypotensive effects. Use with caution.
CNS Depressants (e.g., alcohol, opioids, benzodiazepines)
Increased central nervous system depression, including drowsiness and respiratory depression.
Storage
Store in a cool, dry place, below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include drowsiness, dizziness, nausea, vomiting, muscle weakness, hypothermia, respiratory depression, seizures, and coma. Treatment is primarily supportive, including gastric lavage or induced emesis if recent ingestion, and respiratory support. There is no specific antidote.
Pregnancy & Lactation
Baclofen should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. It is excreted in breast milk in small amounts; therefore, caution should be exercised when administered to a nursing woman. Consult your doctor.
