Overview of the medicine
Caldolin is a combination preparation of Calcium Carbonate and Vitamin D3. It is used to prevent and treat calcium and vitamin D deficiencies, particularly in conditions like osteoporosis, rickets, and during pregnancy/lactation.
Uses & Indications
Dosage
Adults
1-2 tablets daily, or as directed by a physician.
Elderly
1-2 tablets daily, or as directed by a physician. Dosage may need adjustment based on renal function.
Renal_impairment
Use with caution. Monitor serum calcium and phosphate levels. Avoid in severe renal impairment.
How to Take
Orally, preferably with or after meals to enhance calcium absorption and minimize gastric upset. Do not chew or crush the tablet; swallow whole with water.

Calcium carbonate provides elemental calcium essential for bone mineralization, muscle contraction, and nerve function. Vitamin D3 (Cholecalciferol) is crucial for regulating calcium and phosphate homeostasis, facilitating their absorption from the gut and deposition into bones.

Excretion

Absorption
Calcium carbonate is absorbed in the small intestine, requiring gastric acid for dissolution. Vitamin D3 is absorbed from the small intestine, primarily in the jejunum, and requires bile salts.
Metabolism
Calcium is not metabolized. Vitamin D3 is hydroxylated in the liver to 25-hydroxyvitamin D3 and then in the kidneys to the active form, 1,25-dihydroxyvitamin D3 (calcitriol).

Drug Interactions
Levothyroxine

Bisphosphonates

Corticosteroids
Reduce calcium absorption.
Iron supplements
Calcium can reduce iron absorption; separate doses by at least 2 hours.
Thiazide diuretics
May increase the risk of hypercalcemia.
Tetracycline and Quinolone antibiotics
Calcium can interfere with their absorption; separate doses by at least 2-3 hours.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose (hypercalcemia) include nausea, vomiting, constipation, abdominal pain, muscle weakness, excessive thirst, polyuria, drowsiness, and in severe cases, cardiac arrhythmias and coma. Management involves discontinuing the product, hydration, and medical supervision.
Pregnancy & Lactation
Caldolin is generally considered safe and beneficial during pregnancy and lactation for meeting increased calcium and vitamin D requirements. However, always consult a physician before use to ensure appropriate dosing.
