Description
Overview of the medicine
Caloprid 2 mg Tablet contains Prucalopride, a selective serotonin 5-HT4 receptor agonist, used for the symptomatic treatment of chronic constipation in adults when laxatives fail to provide adequate relief.
Uses & Indications
Dosage
Adults
2 mg once daily. Dosage may be reduced to 1 mg once daily in patients with severe renal impairment.
Elderly
Starting dose of 1 mg once daily may be considered in elderly patients, increasing to 2 mg once daily if needed.
Renal_impairment
Severe renal impairment (CrCl < 30 mL/min): 1 mg once daily. No dosage adjustment needed for mild to moderate renal impairment.
How to Take
Oral administration. Can be taken with or without food, at any time of day.
Mechanism of Action
Prucalopride is a selective, high-affinity 5-hydroxytryptamine 4 (5-HT4) receptor agonist. By stimulating 5-HT4 receptors, it enhances colonic motility and accelerates colonic transit, thereby facilitating bowel movements and relieving chronic constipation.
Pharmacokinetics
Onset
Onset of action typically within 2-3 days, but can vary.
Excretion
Primarily excreted via urine (approximately 60% unchanged drug) and feces (approximately 25%).
Half life
Approximately 24-28 hours.
Absorption
Rapidly absorbed after oral administration. Peak plasma concentrations are reached in 2-3 hours. Absolute bioavailability is >90%.
Metabolism
Extensively metabolized to a small extent; the majority is excreted unchanged. Main metabolite is inactive.
