Description
Overview of the medicine
Cephradine is a first-generation cephalosporin antibiotic used to treat various bacterial infections, including those of the respiratory tract, urinary tract, skin and soft tissue, and bones.
Uses & Indications
Dosage
Adults
Usually 250 mg or 500 mg every 6 or 12 hours, depending on the severity and type of infection. Max 4 g/day.
Elderly
No specific dosage adjustment is generally required for elderly patients with normal renal function. However, renal function should be monitored, and dosage adjusted if impaired.
Renal_impairment
Dosage should be reduced in patients with significant renal impairment. Consult product monograph for specific creatinine clearance-based adjustments.
How to Take
Take orally with a glass of water, with or without food. To minimize gastrointestinal upset, it can be taken with food.
Mechanism of Action
Cephradine inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs), leading to bacterial cell lysis and death.
Pharmacokinetics
Onset
Within 30-60 minutes after oral administration.
Excretion
Primarily excreted unchanged in the urine by glomerular filtration and tubular secretion. Approximately 80-90% of the dose is excreted within 6 hours.
Half life
Approximately 0.7-2 hours in adults with normal renal function.
Absorption
Rapidly and well absorbed from the gastrointestinal tract after oral administration. Peak plasma concentrations are achieved within 1-2 hours.
Metabolism
Not significantly metabolized in the body.
Side Effects
Contraindications
- •Hypersensitivity to cephradine or any other cephalosporin antibiotics.
- •History of severe hypersensitivity reactions to penicillin or other beta-lactam antibiotics.
Drug Interactions
Probenecid
May increase cephradine plasma concentrations and prolong its elimination half-life by inhibiting renal tubular secretion.
Aminoglycosides
Concurrent use may increase the risk of nephrotoxicity, especially in patients with pre-existing renal impairment.
Oral Anticoagulants (e.g., Warfarin)
Cephalosporins may enhance the anticoagulant effect; monitor INR/PT.
Storage
Store below 30°C in a dry place, away from direct light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include nausea, vomiting, epigastric distress, diarrhea, and rarely, convulsions. Treatment is supportive and symptomatic. Hemodialysis may be useful in severe cases.
Pregnancy & Lactation
Pregnancy Category B. Studies in animals have not shown harm to the fetus. Use during pregnancy should be only if clearly needed. Cephradine is excreted in low concentrations in breast milk; generally considered compatible with breastfeeding, but caution is advised.
