Description
Overview of the medicine
Ceftibuten is a semi-synthetic, orally administered, third-generation cephalosporin antibiotic. It is used to treat a wide range of bacterial infections sensitive to ceftibuten.
Uses & Indications
Dosage
Adults
400 mg once daily for 5-10 days, depending on the type and severity of infection.
Elderly
No specific dose adjustment is required for elderly patients with normal renal function.
Renal_impairment
For creatinine clearance 30-49 mL/min: 200 mg once daily. For creatinine clearance < 30 mL/min: 100 mg once daily or 400 mg every other day.
How to Take
Ceftibuten capsules can be taken orally with or without food. It is recommended to take it at approximately the same time each day.
Mechanism of Action
Ceftibuten inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs), leading to bactericidal action against susceptible organisms.
Pharmacokinetics
Onset
Clinical effects typically begin within 1-2 hours of administration.
Excretion
Mainly renal; 60-75% of the administered dose is excreted unchanged in the urine over 24 hours.
Half life
Approximately 2-4 hours.
Absorption
Well absorbed orally, bioavailability approximately 75-90%. Peak plasma concentrations are reached within 2-3 hours.
Metabolism
Minimal hepatic metabolism; primarily excreted as unchanged drug.
Side Effects
Contraindications
- Known hypersensitivity to ceftibuten, other cephalosporins, or any component of the formulation.
- Patients with a history of severe hypersensitivity reactions (e.g., anaphylaxis) to any beta-lactam antibiotic.
Drug Interactions
Warfarin and other anticoagulants
May increase anticoagulant effect (INR) in some patients. Monitor INR closely.
Antacids containing aluminum or magnesium
May decrease absorption of ceftibuten. Administer ceftibuten at least 2 hours before or after antacids.
Live bacterial vaccines (e.g., typhoid vaccine)
Antibiotics may reduce the therapeutic effect of live bacterial vaccines. Avoid co-administration.
Storage
Store in a cool, dry place below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
In case of overdose, symptoms are likely to be an exaggeration of known adverse effects. Treatment should be symptomatic and supportive. Hemodialysis may be useful in enhancing the elimination of ceftibuten.
Pregnancy & Lactation
Pregnancy Category B. Studies in animals have shown no evidence of harm to the fetus. However, there are no adequate and well-controlled studies in pregnant women. Use during pregnancy only if clearly needed. Ceftibuten is excreted in low concentrations in human breast milk; use with caution in nursing mothers.
