Description
Overview of the medicine
Celozin 5 mg syrup is an antihistamine used to relieve symptoms of allergies such as sneezing, runny nose, watery eyes, and itching. It contains Cetirizine Hydrochloride, a second-generation H1-receptor antagonist.
Uses & Indications
Dosage
Adults
10 mg (10 ml of syrup) once daily. For some patients, 5 mg (5 ml of syrup) twice daily may be preferred.
Elderly
No dosage adjustment needed for elderly with normal renal function. For renal impairment, dosage should be reduced.
Renal_impairment
Moderate renal impairment (creatinine clearance 31-50 ml/min): 5 mg (5 ml of syrup) once daily. Severe renal impairment (creatinine clearance <31 ml/min): 5 mg (5 ml of syrup) every other day. End-stage renal disease/dialysis patients: Not recommended.
How to Take
Celozin syrup should be taken orally, with or without food. Use a measuring spoon or oral syringe for accurate dosing, especially for children.
Mechanism of Action
Cetirizine selectively inhibits peripheral H1-receptors, thereby preventing the binding of histamine to these receptors and blocking histamine-mediated allergic responses. It has minimal sedative effects due to poor penetration into the central nervous system.
Pharmacokinetics
Onset
Onset of action is typically within 20-60 minutes.
Excretion
Mainly excreted unchanged in the urine (about 70% within 24 hours), with a smaller portion excreted in feces.
Half life
Approximately 8-11 hours in adults; prolonged in children and individuals with renal impairment.
Absorption
Rapidly and extensively absorbed from the gastrointestinal tract after oral administration. Peak plasma concentrations are reached within 1 hour.
Metabolism
Minimal hepatic metabolism; primarily excreted unchanged.
Side Effects
Contraindications
- Hypersensitivity to cetirizine, hydroxyzine, or any component of the formulation.
- Severe renal impairment (creatinine clearance <10 ml/min).
Drug Interactions
Ritonavir
Concurrent use with ritonavir may lead to an increased plasma concentration of cetirizine and/or decreased plasma concentration of ritonavir.
Theophylline
A small decrease in cetirizine clearance has been reported with concurrent use of theophylline (400 mg/day).
Central Nervous System (CNS) Depressants
Concurrent use with alcohol, sedatives, or other CNS depressants may increase drowsiness and impair performance.
Storage
Store below 30°C. Protect from light and moisture. Do not freeze. Keep out of reach of children.
Overdose
Symptoms of overdose may include drowsiness, agitation, headache, tachycardia, tremor, urinary retention, and rarely, convulsions. Management is symptomatic and supportive; there is no specific antidote. Gastric lavage and activated charcoal may be considered if ingestion is recent.
Pregnancy & Lactation
Pregnancy Category B. Studies in animals have shown no evidence of harm to the fetus, but there are no adequate and well-controlled studies in pregnant women. Use only if clearly needed. Cetirizine is excreted into breast milk; therefore, caution should be exercised when administered to a nursing mother.
