Description
Overview of the medicine
Cloxin 500 mg Capsule contains Cloxacillin, a penicillin antibiotic used to treat bacterial infections, particularly those caused by penicillinase-producing staphylococci.
Uses & Indications
Dosage
Adults
250-500 mg every 6 hours, 30 minutes to 1 hour before meals.
Elderly
Similar to adult dosage, but caution with renal impairment. Dose adjustment may be needed.
Renal_impairment
Dose reduction may be necessary in severe renal impairment (e.g., creatinine clearance < 10 mL/min).
How to Take
Take orally with a glass of water, preferably 30 minutes to 1 hour before meals to ensure optimal absorption.
Mechanism of Action
Cloxacillin inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs), leading to bacterial cell lysis and death.
Pharmacokinetics
Onset
Rapid onset of action, typically within 30-60 minutes.
Excretion
Primarily excreted unchanged in the urine, with some biliary excretion.
Half life
Approximately 0.5-1.5 hours.
Absorption
Well absorbed orally, though food may reduce absorption. Peak plasma concentrations are reached within 1-2 hours.
Metabolism
Partially metabolized in the liver to inactive metabolites.
Side Effects
Contraindications
- •Hypersensitivity to penicillin or any component of Cloxacillin
- •History of severe allergic reaction to other beta-lactam antibiotics (e.g., cephalosporins)
Drug Interactions
Warfarin
May enhance anticoagulant effect, requiring closer INR monitoring.
Probenecid
Increases and prolongs plasma levels of Cloxacillin.
Methotrexate
May increase methotrexate toxicity due to reduced renal clearance.
Storage
Store below 30°C in a dry place, protected from light and moisture. Keep out of reach of children.
Overdose
Symptoms may include nausea, vomiting, diarrhea, and convulsions in severe cases. Management is supportive and symptomatic, with gastric lavage or activated charcoal if recent ingestion.
Pregnancy & Lactation
Use in pregnancy only if clearly needed and the potential benefits outweigh the risks. Excreted in breast milk in small amounts; use with caution during lactation, monitoring for adverse effects in the infant.
