Description
Overview of the medicine
Co-Axet 250 mg Tablet is an antibiotic medication containing a combination of Amoxicillin and Clavulanic Acid. It is used to treat a wide range of bacterial infections including respiratory tract infections, urinary tract infections, skin and soft tissue infections, and bone and joint infections. Clavulanic acid helps to prevent bacteria from destroying amoxicillin, making the antibiotic more effective.
Uses & Indications
Dosage
Adults
One Co-Axet 250 mg tablet every 8 hours, or as directed by the physician. Dosage depends on the severity of infection.
Elderly
No special dosage adjustment required unless renal impairment is present. Monitor renal function.
Renal_impairment
Dosage adjustment is necessary based on creatinine clearance. For CrCl 10-30 mL/min, 250 mg every 12 hours. For CrCl <10 mL/min, 250 mg every 24 hours.
How to Take
Take the tablet orally with a glass of water, preferably at the start of a meal to minimize potential gastrointestinal intolerance. Do not crush or chew the tablet.
Mechanism of Action
Amoxicillin is a penicillin antibiotic that inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs), leading to bacterial cell lysis and death. Clavulanic acid is a beta-lactamase inhibitor that inactivates various beta-lactamase enzymes produced by bacteria, which would otherwise destroy amoxicillin, thus extending amoxicillin's antibacterial spectrum.

Absorption
Both amoxicillin and clavulanic acid are well absorbed from the gastrointestinal tract after oral administration. Absorption is not significantly affected by food.
Metabolism
Amoxicillin is minimally metabolized; Clavulanic acid is extensively metabolized.
Side Effects
Contraindications
- •History of severe hypersensitivity reaction to amoxicillin, clavulanic acid, or any penicillin
- •History of cholestatic jaundice/hepatic dysfunction associated with co-amoxiclav therapy
- •Infectious mononucleosis
Drug Interactions
Allopurinol
Concomitant use may increase the incidence of skin rash.
Methotrexate
Co-administration may increase methotrexate toxicity due to reduced renal clearance.
Oral contraceptives
May reduce the effectiveness of oral contraceptives.
Warfarin and other oral anticoagulants
May prolong prothrombin time (INR).
Storage
Store below 30°C in a dry place, away from direct sunlight and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include gastrointestinal symptoms (nausea, vomiting, diarrhea) and disturbances of fluid and electrolyte balance. In rare cases, renal failure and convulsions may occur. Treatment is symptomatic and supportive. Hemodialysis may be useful.
Pregnancy & Lactation
Pregnancy Category B. Use only if clearly needed. Both amoxicillin and clavulanic acid are excreted into breast milk. Use with caution during breastfeeding, monitoring for potential adverse effects in the infant (e.g., diarrhea, candidiasis).
