Description
Overview of the medicine
Cozid 1 gm Injection contains Ceftriaxone, a broad-spectrum third-generation cephalosporin antibiotic, used to treat various bacterial infections.
Uses & Indications
Dosage
Adults
Usual adult dose is 1-2 gm once daily (IM or IV), up to 4 gm daily for severe infections.
Elderly
No dose adjustment typically required unless severe renal or hepatic impairment.
Renal_impairment
No dose adjustment needed for mild to moderate impairment. For severe impairment (CrCl <10 mL/min), maximum 2 gm daily.
How to Take
Can be administered intramuscularly (IM) or intravenously (IV). For IM, reconstitute with lidocaine. For IV, reconstitute with sterile water for injection and administer slowly over 20-30 minutes.
Mechanism of Action
Ceftriaxone inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs), leading to bactericidal activity.
Pharmacokinetics
Onset
Rapid, within minutes for IV, 2-3 hours for IM.
Excretion
Approximately 50-60% excreted unchanged in urine, 40-50% excreted via bile and feces.
Half life
Approximately 6-9 hours.
Absorption
Rapid and complete absorption after intramuscular administration. IV administration provides immediate bioavailability.
Metabolism
Partially metabolized in the liver to inactive metabolites.
Side Effects
Contraindications
- •Hypersensitivity to ceftriaxone or any other cephalosporins
- •Neonates (especially premature) with hyperbilirubinemia
- •Concomitant use with calcium-containing IV solutions in neonates.
Drug Interactions
Warfarin
May increase anticoagulant effect, monitor INR.
Aminoglycosides
Enhanced nephrotoxicity potential, monitor renal function.
Oral contraceptives
May reduce efficacy, advise alternative contraception.
Storage
Store below 30°C in a dry place, away from light. Do not freeze. Reconstituted solution should be used immediately or within 6 hours if stored at room temperature, or 24 hours if refrigerated.
Overdose
In case of overdose, symptomatic and supportive treatment should be given. Ceftriaxone is not significantly removed by hemodialysis or peritoneal dialysis.
Pregnancy & Lactation
Pregnancy category B. Use with caution during pregnancy if clearly needed. Excreted in low concentrations in breast milk, use with caution during lactation.
