Description
Overview of the medicine
Deslosil (Desloratadine) is a non-sedating antihistamine used to relieve symptoms of allergic rhinitis (e.g., sneezing, runny nose, itchy eyes) and chronic idiopathic urticaria (hives).
Uses & Indications
Dosage
Adults
5 mg once daily.
Elderly
No dosage adjustment is generally required, but caution is advised due to potential age-related decrease in renal or hepatic function.
Renal_impairment
For patients with severe renal impairment, a starting dose of 5 mg every other day is recommended.
How to Take
Deslosil tablets should be taken orally, with or without food. Swallow the tablet whole with water.
Mechanism of Action
Desloratadine is a potent, long-acting, selective peripheral H1-receptor antagonist. It inhibits the release of histamine from mast cells and basophils and also blocks various inflammatory mediators like leukotrienes and prostaglandins.

Excretion
Primarily excreted in urine and feces as metabolites.
Half life
Approximately 27 hours, allowing for once-daily dosing.
Absorption
Rapidly absorbed after oral administration. Peak plasma concentrations are reached approximately 3 hours after dosing.
Metabolism
Extensively metabolized in the liver to 3-hydroxydesloratadine, an active metabolite, primarily by CYP2C8 and CYP2D6.

Desloratadine is excreted in breast milk; therefore, a decision should be made whether to discontinue nursing or to discontinue the drug, taking into account the importance of the drug to the mother.
