Don-A 5 mg Suspension is an oral medication containing Domperidone, primarily used to treat nausea and vomiting, and to improve gastric motility.
Uses & Indications
Dosage
Adults
10 mg (10 ml of suspension) three to four times daily, before meals.
Elderly
Similar to adult dosage, but caution due to potential for cardiac side effects. Lower starting dose may be considered.
Renal_impairment
Dosage reduction may be necessary; consider 10 mg once or twice daily depending on severity. Monitor closely.
How to Take
Oral use. Should be taken 15-30 minutes before meals. Do not exceed the recommended dose.
Mechanism of Action
Domperidone acts as a peripheral dopamine D2 receptor antagonist, primarily in the chemoreceptor trigger zone (CTZ) and the gastrointestinal tract. This action blocks dopamine-induced emesis and enhances gastrointestinal motility.

Side Effects
Contraindications

- •Patients with known prolongation of cardiac QTc intervals
- •Co-administration with potent CYP3A4 inhibitors (e.g., ketoconazole, erythromycin)
Drug Interactions
Anticholinergics
May antagonize the prokinetic effect of domperidone.
QTc-prolonging drugs
Increased risk of cardiac arrhythmias. Concomitant use is contraindicated or requires extreme caution.
Potent CYP3A4 inhibitors
Increased plasma levels of domperidone, increased risk of QTc prolongation. Concomitant use is contraindicated.
Storage
Store below 30°C, protected from light and moisture. Do not freeze.
Overdose
Symptoms of overdose include drowsiness, disorientation, extrapyramidal reactions, and potential cardiac effects. Management is symptomatic and supportive, including gastric lavage and activated charcoal. ECG monitoring is recommended.
Pregnancy & Lactation
Pregnancy Category C. Use during pregnancy only if clearly needed and the potential benefits outweigh the risks. Domperidone is excreted in breast milk; therefore, use is generally not recommended during lactation, especially if cardiac risks for the infant are a concern.
