Overview of the medicine
Doxolator 400 mg Tablet contains Doxifloxacin, a broad-spectrum fluoroquinolone antibiotic used to treat a variety of bacterial infections.
Uses & Indications
Dosage
Adults
400 mg once daily for 5-10 days, depending on the type and severity of infection. Duration may vary.
Elderly
No specific dose adjustment required based on age alone; however, renal function should be assessed and dose adjusted if needed.
Renal_impairment
For creatinine clearance <30 mL/min, the dose should be reduced (e.g., 200 mg once daily or 400 mg every 48 hours).
How to Take
Take orally, with or without food. Swallow the tablet whole with a glass of water. Do not crush or chew.
Mechanism of Action
Doxifloxacin exerts its bactericidal effect by inhibiting bacterial DNA gyrase and topoisomerase IV, essential enzymes for bacterial DNA replication, transcription, repair, and recombination.

Primarily excreted unchanged in the urine, with some fecal excretion.

Approximately 8-12 hours, allowing for once-daily dosing.
Absorption
Rapidly and well absorbed from the gastrointestinal tract after oral administration, with peak plasma concentrations reached within 1-2 hours.
Metabolism
Minimally metabolized in the liver.
Side Effects
Contraindications
- Hypersensitivity to Doxifloxacin or other fluoroquinolones
- History of tendon disorders related to fluoroquinolone use
- Children and adolescents under 18 years of age (due to risk of arthropathy)
Drug Interactions
Warfarin
May enhance the anticoagulant effect of warfarin, requiring monitoring of INR.
Corticosteroids
Increased risk of tendon rupture with concomitant use.
NSAIDs (Nonsteroidal Anti-inflammatory Drugs)
Concurrent use may increase the risk of CNS stimulation and convulsive seizures.
Antacids (containing aluminum, magnesium, or calcium)
May significantly decrease Doxifloxacin absorption. Administer Doxifloxacin at least 2 hours before or 4 hours after antacids.
Storage
Store below 30°C in a dry place, away from direct sunlight and moisture. Keep out of reach of children.

In case of overdose, symptoms may include nausea, vomiting, diarrhea, and central nervous system effects. Management is primarily symptomatic and supportive, including gastric lavage and hydration. Hemodialysis is not effective in removing Doxifloxacin.
Pregnancy & Lactation
Not recommended during pregnancy due to potential harm to the fetus. Use during lactation is generally discouraged as Doxifloxacin passes into breast milk and may harm the infant. Consult a doctor.
