Description
Overview of the medicine
Doxonil 100 mg Syrup contains Doxofylline, a xanthine derivative bronchodilator. It is used for the treatment of bronchial asthma, COPD, and other bronchospastic conditions. It works by relaxing the smooth muscles of the airways, making breathing easier.
Uses & Indications
Dosage
Adults
10-20 ml (200-400 mg) twice daily, or as directed by the physician. Maximum 1200 mg/day.
Elderly
Dosage adjustment may be required based on individual response and renal/hepatic function. Typically, lower doses are recommended, e.g., 10 ml (200 mg) twice daily.
Renal_impairment
Caution advised. Dosage reduction may be necessary. Close monitoring of renal function is recommended.
How to Take
Oral administration. Take with or without food. Use a measuring spoon or cup for accurate dosing. Shake well before use.
Mechanism of Action
Doxofylline is a bronchodilator that inhibits phosphodiesterase enzymes, leading to increased intracellular cAMP levels. This causes relaxation of bronchial smooth muscles, resulting in bronchodilation. It also has anti-inflammatory properties.
Pharmacokinetics
Onset
Within 30-60 minutes
Excretion
Excreted mainly in the urine as inactive metabolites.
Half life
Approximately 6-8 hours
Absorption
Rapidly absorbed from the gastrointestinal tract. Peak plasma concentrations reached within 1-2 hours.
Metabolism
Primarily metabolized in the liver (over 90%) by oxidation, forming inactive metabolites. It does not undergo extensive first-pass metabolism.
Side Effects
Contraindications
- Hypersensitivity to Doxofylline or other xanthine derivatives
- Acute myocardial infarction
- Severe hypotension
- Lactation
- Children below 6 years
Drug Interactions
Beta-blockers
May reduce the bronchodilatory effect.
Cimetidine, Allopurinol
May inhibit Doxofylline metabolism, increasing its concentration.
Phenytoin, Phenobarbital, Rifampicin
May decrease Doxofylline plasma levels, reducing efficacy.
Erythromycin, Lincomycin, Clindamycin
May increase Doxofylline plasma levels, leading to increased side effects.
Storage
Store below 30°C in a dry place, away from direct light. Do not freeze. Keep out of reach of children.
Overdose
Symptoms may include severe nausea, vomiting, epigastric pain, restlessness, insomnia, headache, tachycardia, arrhythmias, and convulsions. Management involves symptomatic and supportive care. Gastric lavage and activated charcoal may be considered.
Pregnancy & Lactation
Pregnancy Category C. Use only if clearly needed and the potential benefits outweigh the potential risks to the fetus. Contraindicated during lactation as Doxofylline is excreted in breast milk.

Clinical Trials
Extensive clinical trials have shown Doxofylline to be effective as a bronchodilator with a favorable safety profile compared to traditional xanthines, particularly regarding cardiac side effects. Ongoing research explores its anti-inflammatory roles.
Lab Monitoring
- Periodic monitoring of plasma Doxofylline levels may be necessary in some patients, especially those with hepatic or renal impairment, or if toxicity is suspected.
Doctor Notes
- Advise patients to report any signs of toxicity (e.g., severe nausea, convulsions).
- Monitor for potential drug interactions, especially with other xanthines or medications affecting hepatic metabolism.
- Ensure proper education on dose measurement for syrup to avoid under or overdose.
Patient Guidelines
- Take medicine as prescribed by your doctor.
- Do not stop taking the medicine without consulting your doctor.
- Inform your doctor about all other medications you are taking.
- Report any severe side effects immediately.
Missed Dose Advice
If you miss a dose, take it as soon as you remember. If it is almost time for your next dose, skip the missed dose and resume your regular dosing schedule. Do not double the dose to catch up.
Driving Precautions
Doxofylline may cause dizziness or visual disturbances in some individuals. If you experience these effects, avoid driving or operating machinery.
Lifestyle Advice
- Avoid smoking as it can reduce the effectiveness of Doxofylline.
- Maintain a healthy lifestyle with regular exercise and a balanced diet.
- Avoid exposure to allergens and irritants that trigger asthma or COPD.
