Description
Overview of the medicine
Ebastine is a non-sedating antihistamine used for the symptomatic treatment of allergic rhinitis (seasonal and perennial) and chronic idiopathic urticaria.
Uses & Indications
Dosage
Adults
10 mg once daily. In severe cases, the dose may be increased to 20 mg once daily.
Elderly
No dose adjustment is generally required, but caution is advised in patients with severe renal or hepatic impairment.
Renal_impairment
No dose adjustment for mild to moderate renal impairment. Caution and dose reduction may be needed in severe renal impairment.
How to Take
Ebahist 10 mg Tablet should be taken orally, with or without food. It should be swallowed whole with water.
Mechanism of Action
Ebastine is a selective peripheral H1-receptor antagonist. It works by blocking the action of histamine, a substance in the body that causes allergic symptoms.
Pharmacokinetics
Onset
Onset of action within 1-3 hours.
Excretion
Mainly excreted in urine (60-70%) as metabolites.
Half life
Parent drug: 3-6 hours; Active metabolite (carebastine): 15-19 hours.
Absorption
Rapidly absorbed after oral administration and extensively metabolized to its active metabolite, carebastine. Bioavailability is approximately 60%.
Metabolism
Extensively metabolized in the liver to carebastine (active metabolite) primarily via CYP3A4.
Side Effects
Contraindications
- •Hypersensitivity to ebastine or any component of the formulation.
- •Children under 12 years of age (for 10mg tablet formulation).
Drug Interactions
Rifampicin
May decrease plasma concentrations of ebastine due to enzyme induction.
Grapefruit juice
Concomitant use with grapefruit juice may increase plasma levels of ebastine.
Ketoconazole, Itraconazole, Erythromycin
May increase plasma levels of ebastine and its active metabolite carebastine, potentially increasing the risk of QTc prolongation.
Storage
Store below 30°C, in a dry place, protected from light and moisture. Keep out of reach of children.
Overdose
In case of overdose, symptomatic and supportive treatment should be initiated. There is no specific antidote. ECG monitoring is recommended.
Pregnancy & Lactation
Pregnancy Category B. Use during pregnancy only if clearly needed. Ebastine is excreted in breast milk; therefore, it should not be used during breastfeeding.
