Overview of the medicine
Edenil 20 mg Tablet contains Esomeprazole, a proton pump inhibitor used to reduce stomach acid production. It is primarily used to treat conditions like gastroesophageal reflux disease (GERD), peptic ulcers, and Zollinger-Ellison syndrome.
Uses & Indications
Dosage
Adults
GERD: 20-40 mg once daily for 4-8 weeks. Erosive Esophagitis: 20-40 mg once daily for 4-8 weeks. PUD: 20 mg once daily for 4-8 weeks. Zollinger-Ellison Syndrome: Initial dose 40 mg twice daily, adjusted as needed.
Elderly
No dose adjustment generally required, but caution advised due to potential age-related decrease in hepatic function.
Renal_impairment
No dose adjustment required for mild to moderate renal impairment. For severe impairment, caution is advised.
How to Take
Take the tablet whole, swallowed with water, at least one hour before a meal. Do not chew or crush. For patients unable to swallow, tablets can be dispersed in non-carbonated water or apple juice.
Mechanism of Action
Esomeprazole is an irreversible proton pump inhibitor that blocks the H+/K+-ATPase enzyme system (the proton pump) in the gastric parietal cells, thereby inhibiting the final step in gastric acid secretion.

Mainly excreted in urine (about 80%) and feces (about 20%) as metabolites.

Absorption
Rapidly absorbed from the small intestine after oral administration. Bioavailability is approximately 64% after a single 40 mg dose and increases to 89% with repeated once-daily administration.
Metabolism
Extensively metabolized in the liver by the cytochrome P450 (CYP) enzyme system, mainly CYP2C19 and CYP3A4.
Side Effects
Contraindications
- Hypersensitivity to Esomeprazole, substituted benzimidazoles, or any component of the formulation
- Concomitant use with nelfinavir
Drug Interactions
Diazepam
Reduced metabolism and increased plasma concentration of diazepam.
Warfarin
Increased INR and prothrombin time.
Clopidogrel
Reduced antiplatelet effect of clopidogrel (CYP2C19 inhibition).
Methotrexate
Possible increase in methotrexate levels.
Ketoconazole/Itraconazole
Reduced absorption due to increased gastric pH.

Store in a cool, dry place, below 30°C. Protect from light and moisture. Keep out of reach of children.

Pregnancy Category B. Studies in animals have not shown harm to the fetus, but well-controlled studies in pregnant women are lacking. Use only if clearly needed. Excretion into breast milk is unknown; caution advised during lactation.
