Overview of the medicine
Edenil 40 mg Tablet contains Esomeprazole, which is used to reduce the amount of acid produced in the stomach. It treats conditions caused by too much stomach acid.
Uses & Indications
Dosage
Adults
20 mg or 40 mg once daily, depending on the indication and severity. For erosive esophagitis, typically 40 mg once daily for 4-8 weeks. For GERD symptoms, 20 mg once daily.
Elderly
No dosage adjustment is generally required for elderly patients.
Renal_impairment
No dosage adjustment is necessary for patients with impaired renal function.
How to Take
Take Edenil 40 mg Tablet orally, usually once daily, at least one hour before a meal. Do not crush, chew, or split the tablet; swallow it whole with water.
Mechanism of Action
Esomeprazole is a proton pump inhibitor that works by irreversibly blocking the H+/K+-ATPase enzyme system (the proton pump) in the gastric parietal cells. This action inhibits the final step in gastric acid production, leading to a reduction in both basal and stimulated acid secretion.
Pharmacokinetics
Onset
Acid suppression occurs within 1 hour after a single oral dose.
Excretion
Approximately 80% of a dose is excreted as metabolites in the urine, and the remainder in the feces.
Half life
The plasma elimination half-life is approximately 1-1.5 hours.
Absorption
Rapidly absorbed after oral administration. Bioavailability is approximately 50-60% after a single 40 mg dose, increasing with repeated dosing.
Metabolism
Extensively metabolized in the liver by the cytochrome P450 (CYP) enzyme system, primarily by CYP2C19 and CYP3A4.
Side Effects
Contraindications
- Hypersensitivity to esomeprazole, substituted benzimidazoles, or any component of the formulation.
- Concomitant use with nelfinavir.
Drug Interactions
Warfarin
Increased INR and prothrombin time.
Clopidogrel
Reduced antiplatelet efficacy of clopidogrel.
Methotrexate
Increased methotrexate levels.
Nelfinavir/Atazanavir
Reduced plasma levels of nelfinavir and atazanavir, leading to loss of virologic response.

Store in a cool, dry place, below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Limited experience with overdose. Symptoms can include drowsiness, confusion, blurred vision, tachycardia, nausea, sweating, headache, dry mouth. Treatment is symptomatic and supportive.
Pregnancy & Lactation
Pregnancy Category C. Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. Esomeprazole is excreted in human milk; caution should be exercised when administered to a nursing woman.
