Description
Overview of the medicine
Etodolac is a non-steroidal anti-inflammatory drug (NSAID) used for the treatment of pain, osteoarthritis, and rheumatoid arthritis. It provides relief by reducing inflammation and pain.
Uses & Indications
Dosage
Adults
For osteoarthritis and rheumatoid arthritis, 300 mg two or three times daily, or 400 mg to 500 mg twice daily, or 600 mg once daily (SR). For acute pain, 200-400 mg every 6-8 hours as needed, not exceeding 1200 mg/day.
Elderly
Start with the lowest effective dose and monitor for adverse effects, especially gastrointestinal bleeding and renal impairment. Dosage adjustment may be necessary.
Renal_impairment
Use with caution in patients with mild to moderate renal impairment. Avoid in severe renal impairment.
How to Take
Take orally with food or milk to minimize gastrointestinal upset. Swallow whole; do not crush or chew sustained-release tablets.
Mechanism of Action
Etodolac exerts its anti-inflammatory, analgesic, and antipyretic effects primarily through the inhibition of prostaglandin synthesis by inhibiting cyclooxygenase (COX-1 and COX-2) enzymes. This reduces the formation of prostaglandins, which are mediators of inflammation and pain.

Onset
Pain relief typically begins within 30-60 minutes.
Excretion
Primarily excreted in the urine (about 60% as metabolites and less than 1% as unchanged drug), and a small amount in feces.
Half life
Approximately 6-7 hours.
Absorption
Well absorbed from the gastrointestinal tract, bioavailability is about 80%.
Metabolism
Extensively metabolized in the liver, primarily by hydroxylation, glucuronidation, and oxidation.
Side Effects
Contraindications
- •Hypersensitivity to Etodolac or any other NSAID
- •History of asthma, urticaria, or allergic-type reactions after taking aspirin or other NSAIDs
- •Peri-operative pain in the setting of coronary artery bypass graft (CABG) surgery
- •Active gastrointestinal bleeding or peptic ulcer disease
- •Severe heart failure, renal or hepatic impairment
Drug Interactions
Lithium
Increased plasma lithium levels.
Methotrexate
Increased methotrexate toxicity.
Ciclosporin/Tacrolimus
Increased nephrotoxicity.
Warfarin/Anticoagulants
Increased risk of bleeding.
ACE inhibitors/Diuretics
Reduced antihypertensive and diuretic effects, increased risk of renal impairment.
Storage
Store in a cool and dry place, below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms may include lethargy, drowsiness, nausea, vomiting, epigastric pain, and gastrointestinal bleeding. Management involves supportive and symptomatic treatment, maintaining airway, monitoring vital signs, and gastric lavage or activated charcoal if recent ingestion.
Pregnancy & Lactation
Pregnancy Category C (D in 3rd trimester or near delivery). Use during pregnancy should be avoided, especially in the third trimester due to potential premature closure of the fetal ductus arteriosus. Not recommended during breastfeeding as it is excreted in breast milk.
