Description
Overview of the medicine
Eziflo is an alpha-blocker used to treat the symptoms of an enlarged prostate (benign prostatic hyperplasia or BPH). It helps to relax the muscles in the prostate and bladder neck, making it easier to urinate.
Uses & Indications
Dosage
Adults
0.4 mg orally once daily, approximately 30 minutes after the same meal each day. The dose may be increased to 0.8 mg once daily after 2 to 4 weeks if necessary.
Elderly
No dosage adjustment is necessary for elderly patients.
Renal_impairment
No dosage adjustment is necessary for patients with mild to moderate renal impairment. Caution is advised in patients with severe renal impairment (CrCl < 10 mL/min).
How to Take
Take orally, as a whole capsule, approximately 30 minutes after the same meal each day. Do not crush, chew, or open the capsule.
Mechanism of Action
Tamsulosin selectively blocks alpha-1A adrenoceptors in the prostate, bladder neck, and prostatic urethra. This blockade causes relaxation of smooth muscles in these areas, reducing resistance to urine flow and improving symptoms of BPH.
Pharmacokinetics
Onset
Clinical effects may be noticeable within 4-6 hours, but optimal symptomatic relief may take several weeks.
Excretion
Primarily excreted in the urine (76%) and feces (21%) mainly as metabolites.
Half life
Approximately 9 to 13 hours in healthy volunteers and 14 to 15 hours in the target population.
Absorption
Well absorbed from the gastrointestinal tract, nearly 90% bioavailability under fed conditions. Peak plasma concentrations are reached about 6 hours after dosing under fed conditions.
Metabolism
Extensively metabolized by cytochrome P450 enzymes in the liver, primarily CYP3A4 and CYP2D6.
Side Effects
Contraindications
- •Hypersensitivity to Tamsulosin or any component of the formulation.
- •History of orthostatic hypotension.
- •Severe hepatic impairment.
Drug Interactions
Warfarin
Limited data, but caution advised due to potential for increased bleeding.
Other alpha-1 blockers
Increased risk of hypotensive effects.
Strong CYP2D6 inhibitors (e.g., paroxetine)
May increase tamsulosin exposure. Use with caution.
PDE5 inhibitors (e.g., sildenafil, tadalafil)
May lead to additive hypotensive effects; use with caution.
Strong CYP3A4 inhibitors (e.g., ketoconazole, ritonavir)
May increase tamsulosin exposure. Use with caution.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Overdose may lead to severe hypotensive effects. Management involves supportive measures, including maintaining cardiovascular stability, fluid replacement, and vasopressors if necessary. Patient should be kept in a supine position.
Pregnancy & Lactation
Eziflo is not indicated for use in women. It is contraindicated in pregnancy and lactation.
