How to Take
Administer orally using the calibrated dropper provided. Do not exceed the recommended dose and dosing interval. Can be given with or without food.
Mechanism of Action
Paracetamol exerts its analgesic and antipyretic effects by inhibiting prostaglandin synthesis, primarily in the central nervous system.
Pharmacokinetics
Onset
Analgesic effect begins within 30-60 minutes, antipyretic effect within 30 minutes.
Excretion
Mainly excreted in the urine as glucuronide and sulfate conjugates. Less than 5% is excreted unchanged.
Half life
Plasma elimination half-life is 1-4 hours (typically 2-3 hours) in healthy individuals.
Absorption
Rapidly and almost completely absorbed from the gastrointestinal tract, with peak plasma concentrations occurring within 30-60 minutes.
Metabolism
Primarily metabolized in the liver via glucuronidation and sulfation, and a small amount via cytochrome P450 pathway to a toxic intermediate (NAPQI) which is rapidly detoxified by glutathione.
Side Effects
Contraindications
- Hypersensitivity to paracetamol or any component of the formulation
- Severe hepatic impairment or active liver disease
Drug Interactions
Warfarin
May increase the anticoagulant effect, leading to a higher risk of bleeding, especially with prolonged daily use.
Cholestyramine
May decrease the absorption of paracetamol if given within one hour.
Metoclopramide, Domperidone
May increase the absorption rate of paracetamol.
Carbamazepine, Phenobarbital, Phenytoin
May increase the risk of hepatotoxicity due to enzyme induction.
Storage
Store below 30°C. Protect from light and moisture. Do not freeze. Keep the bottle tightly closed.
Overdose
Symptoms of overdose include nausea, vomiting, abdominal pain, pallor, and sweating. Severe overdose can lead to hepatic necrosis and renal tubular necrosis. Treatment involves gastric lavage, activated charcoal, and administration of N-acetylcysteine (NAC) as an antidote.
Pregnancy & Lactation
While this is a pediatric formulation, paracetamol is generally considered safe for use during pregnancy and lactation when used at recommended doses. Consult a healthcare professional before use.
