Description
Overview of the medicine
Flunapen 50 mg Capsule contains Flucloxacillin, an antibiotic belonging to the penicillin group. It is used to treat various bacterial infections, especially those caused by penicillinase-producing staphylococci. The 50 mg strength is typically for pediatric use or very specific mild infections.
Uses & Indications
Dosage
Adults
The typical adult dose of Flucloxacillin is 250-500 mg every 6 hours. For Flunapen 50 mg, the dosage would be specifically determined by the physician based on the infection severity and patient's condition, likely for very mild infections or as a pediatric dose.
Elderly
No specific dose reduction required unless severe renal impairment. Regular adult dose may be used.
Renal_impairment
Dose adjustment required for severe renal impairment (creatinine clearance <10 mL/min). A reduction in frequency or dose may be necessary. For mild to moderate impairment, no change.
How to Take
Oral administration. Take on an empty stomach (at least 30-60 minutes before food or 2 hours after food) for optimal absorption. Swallow the capsule whole with water.
Mechanism of Action
Flucloxacillin inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins (PBPs), leading to bacterial cell lysis and death.
Pharmacokinetics
Onset
Rapid onset, typically within 30-60 minutes.
Excretion
Mainly excreted unchanged in the urine, with some biliary excretion.
Half life
Approximately 0.5 to 1 hour.
Absorption
Well absorbed from the gastrointestinal tract, though food may reduce absorption. Peak plasma concentrations usually occur 1 hour after administration.
Metabolism
Partially metabolized in the liver to inactive metabolites.
Side Effects
Contraindications
- •Hypersensitivity to penicillin antibiotics or any component of the formulation
- •History of flucloxacillin-associated jaundice/hepatic dysfunction
Drug Interactions
Warfarin
May enhance the anticoagulant effect of warfarin, requiring close monitoring of INR.
Probenecid
Increases and prolongs plasma levels of flucloxacillin by competing for renal tubular secretion.
Methotrexate
May increase methotrexate plasma concentrations, enhancing its toxicity.
Oral Contraceptives
May reduce the effectiveness of oral contraceptives (though clinical significance is debated, advise additional contraception).
Storage
Store below 25°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include nausea, vomiting, diarrhea, and convulsions. Management is symptomatic and supportive, with no specific antidote. Gastric lavage or activated charcoal may be considered if recent ingestion.
Pregnancy & Lactation
Pregnancy Category B. Generally considered safe during pregnancy, but use only if clearly needed. Flucloxacillin is excreted in breast milk; caution advised during lactation.
