Fluzole 50 mg Tablet contains Fluconazole, an antifungal medication used to treat a variety of fungal and yeast infections. It belongs to the azole class of antifungals and works by inhibiting the growth of fungi. It is effective against candidiasis, cryptococcosis, and other susceptible fungal infections.
Uses & Indications
Dosage
Adults
Vaginal candidiasis: 150 mg single oral dose. Oropharyngeal candidiasis: 50-100 mg once daily for 7-14 days. Esophageal candidiasis: 100-200 mg once daily for 14-30 days. For other systemic infections, dosage may vary, usually 100-400 mg daily.
Elderly
No dosage adjustment is typically required unless there is renal impairment. Dose should be adjusted based on creatinine clearance.
Renal_impairment
For patients with creatinine clearance < 50 mL/min, the dose should be reduced by 50% after an initial loading dose.
How to Take
Fluzole tablets should be swallowed whole with water, with or without food. For oral candidiasis, swishing and swallowing the liquid suspension may be preferred for better local action, but tablets are generally swallowed. Complete the full course of treatment as prescribed.
Mechanism of Action
Fluconazole inhibits the fungal cytochrome P450 enzyme 14α-demethylase, which is responsible for converting lanosterol to ergosterol. Ergosterol is an essential component of the fungal cell membrane; its inhibition leads to increased cellular permeability and leakage of cellular contents, ultimately causing fungal cell death.

Within 1-2 days for most infections, a single dose for vaginal candidiasis can show improvement within 24 hours.
Excretion
Primarily excreted unchanged by the kidneys (approx. 80% of the dose).
Half life
Approximately 30 hours, allowing for once-daily dosing in many indications.
Absorption
Well absorbed orally (over 90% bioavailability); food does not significantly affect absorption.
Metabolism
Minimally metabolized; about 11% of the dose is metabolized to less active metabolites.
Side Effects
Contraindications
- Hypersensitivity to Fluconazole or other azole antifungals.
- Co-administration with terfenadine, astemizole, cisapride, pimozide, quinidine, or erythromycin in patients receiving multiple doses of fluconazole.
Drug Interactions
Warfarin
Increased prothrombin time, monitor INR.
Phenytoin
Increased phenytoin levels, monitor for toxicity.
Rifampicin
Decreased fluconazole levels, consider increasing fluconazole dose.
Cyclosporine, Tacrolimus
Increased levels of immunosuppressants, monitor levels.
Oral Hypoglycemics (e.g., sulfonylureas)
Increased blood glucose lowering effect, monitor blood sugar.

In case of overdose, symptoms may include hallucinations and paranoid behavior. Management should be symptomatic and supportive, including gastric lavage if appropriate and hemodialysis, which significantly decreases plasma levels.

Pregnancy Category D for chronic use or high doses; Category C for single-dose vaginal candidiasis. Avoid during pregnancy unless the benefits outweigh the risks. Excreted into breast milk; caution is advised during breastfeeding.
