Description
Overview of the medicine
Fritt 250 mg Tablet contains Amoxicillin, a penicillin-type antibiotic used to treat a wide variety of bacterial infections. It works by stopping the growth of bacteria. This antibiotic treats only bacterial infections and will not work for viral infections (such as common cold, flu).
Uses & Indications
Dosage
Adults
Standard adult dose is 250 mg to 500 mg every 8 hours, or 500 mg to 875 mg every 12 hours, depending on the severity and type of infection. For severe infections, doses may be increased. Max: 4 g/day.
Elderly
No specific dose adjustment is generally required for elderly patients with normal renal function. Dose adjustments may be necessary based on renal function.
Renal_impairment
Dose reduction is required for patients with severe renal impairment (CrCl < 30 mL/min). A common recommendation is 250 mg every 8-12 hours, depending on the severity.
How to Take
Fritt 250 mg Tablet should be taken orally, with or without food. For best absorption and to minimize gastrointestinal upset, it can be taken at the start of a meal. Swallow the tablet whole with a glass of water. Do not chew or crush it.
Mechanism of Action
Amoxicillin works by inhibiting the biosynthesis of bacterial cell wall peptidoglycan. It binds to penicillin-binding proteins (PBPs) located in the bacterial cell wall, which prevents the cross-linking of peptidoglycan polymers, leading to cell lysis and bacterial death.
Pharmacokinetics
Onset
Within 30-60 minutes.
Excretion
Primarily excreted unchanged in the urine via renal tubular secretion and glomerular filtration. About 60-80% of an oral dose is excreted in the urine within 6-8 hours.
Half life
Approximately 1-1.5 hours in adults with normal renal function.
Absorption
Rapidly and well absorbed from the gastrointestinal tract, with bioavailability of approximately 70-90%. Peak plasma concentrations are reached within 1-2 hours after oral administration.
Metabolism
Limited metabolism, mainly converted to inactive penicilloic acid.
Side Effects
Contraindications
- •Known hypersensitivity to penicillin or any component of the formulation.
- •Patients with infectious mononucleosis (increased risk of rash).
Drug Interactions
Probenecid
Increases and prolongs plasma levels of amoxicillin by decreasing renal tubular secretion.
Allopurinol
Increased incidence of rash, particularly in patients with hyperuricemia.
Methotrexate
Increased methotrexate toxicity due to reduced renal excretion. Monitor methotrexate levels.
Oral Contraceptives
May reduce efficacy of oral contraceptives (rarely). Advise additional contraceptive methods.
Oral Anticoagulants (e.g., Warfarin)
Increased INR/bleeding risk. Monitor coagulation parameters.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
In case of overdose, gastrointestinal symptoms such as nausea, vomiting, and diarrhea may occur. Renal impairment leading to crystalluria has been reported in a few cases. Treatment is largely symptomatic and supportive. Hemodialysis may be useful in removing Amoxicillin from circulation.
Pregnancy & Lactation
Pregnancy Category B. Studies in animals have shown no evidence of harm to the fetus. Amoxicillin is generally considered safe during pregnancy when clearly needed. It is excreted in breast milk in small amounts, but is generally considered compatible with breastfeeding. Consult a doctor before use during pregnancy or lactation.
