Fungitrol 50 mg Suspension, containing Fluconazole, is an antifungal medication used to treat a variety of fungal infections, including candidiasis and cryptococcal meningitis. It works by inhibiting the growth of fungi.
Uses & Indications
Dosage
Adults
Varies by indication; e.g., 50-400 mg once daily. For oral candidiasis, 50-100 mg once daily for 7-14 days. For vaginal candidiasis, 150 mg single dose.
Elderly
Dose adjustment may be needed based on renal function (creatinine clearance).
Renal_impairment
For creatinine clearance < 50 mL/min, the dose should be reduced by 50% after the initial loading dose.
How to Take
Oral administration. Shake well before use. Can be taken with or without food.
Mechanism of Action
Fluconazole inhibits fungal cytochrome P450-dependent 14-alpha-demethylase, which is essential for ergosterol biosynthesis. This inhibition leads to a depletion of ergosterol and a corresponding accumulation of 14-alpha-methyl sterols in the fungal cell, resulting in membrane disruption and inhibition of fungal growth.

Side Effects
Contraindications
- •Hypersensitivity to fluconazole or other azole antifungal agents.
- •Co-administration with drugs known to prolong the QT interval and metabolized by CYP3A4, such as terfenadine or cisapride.
Drug Interactions
Warfarin
Increased prothrombin time (INR) and risk of bleeding.
Phenytoin
Increases phenytoin plasma concentrations.
Rifampicin
Decreases fluconazole plasma concentrations.
Cyclosporine, Tacrolimus
Increases plasma concentrations of these immunosuppressants.
Oral Hypoglycemic Agents
Prolongs the half-life of oral sulfonylureas, increasing risk of hypoglycemia.
Storage
Store below 25°C in a dry place, protected from light and moisture. Keep out of reach of children.
Overdose
In the event of an overdose, symptomatic treatment and supportive measures should be implemented. Gastric lavage may be considered. Hemodialysis can reduce plasma concentrations significantly.
Pregnancy & Lactation
Pregnancy Category C. Should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus. Fluconazole is excreted in breast milk; caution should be exercised when administered to a nursing woman.
