Description
Overview of the medicine
G-Calbo DX is a combination supplement of Calcium Carbonate and Vitamin D3 (Cholecalciferol). It is used to prevent and treat calcium and vitamin D deficiencies, promote bone health, and support various bodily functions.
Uses & Indications
Dosage
Adults
1-2 tablets daily, or as directed by a physician. Best taken with or after meals.
Elderly
Same as adults, but caution should be exercised in patients with reduced renal function.
Renal_impairment
Use with caution. Dose adjustment may be necessary. Regular monitoring of serum calcium and phosphate levels is recommended.
How to Take
Oral administration. Tablets should be swallowed with water, preferably with or after meals to enhance absorption and reduce gastric upset.
Mechanism of Action
Calcium is essential for building and maintaining strong bones, nerve transmission, muscle contraction, and blood clotting. Vitamin D3 facilitates the absorption of calcium from the intestine, regulates serum calcium and phosphate levels, and promotes bone mineralization.
Pharmacokinetics
Onset
The therapeutic effects on bone health are gradual, typically becoming evident after several weeks to months of consistent use.
Excretion
Calcium is primarily excreted via the kidneys (urine) and the gastrointestinal tract (feces). Vitamin D3 metabolites are mainly excreted in bile and feces.
Half life
Calcium has complex pharmacokinetics; its half-life varies. Vitamin D3's half-life is approximately 24 hours, but its active metabolites have longer half-lives (e.g., 25-hydroxyvitamin D3 has a half-life of 2-3 weeks).
Absorption
Calcium is absorbed primarily in the small intestine via active transport and passive diffusion. Vitamin D3 is absorbed from the small intestine, requiring bile salts for efficient absorption.
Metabolism
Calcium is not metabolized. Vitamin D3 is hydroxylated in the liver to 25-hydroxyvitamin D3 (calcifediol) and then in the kidneys to 1,25-dihydroxyvitamin D3 (calcitriol), its active form.
Side Effects
Contraindications
- •Hypercalcemia (abnormally high levels of calcium in the blood)
- •Hypercalciuria (excessive excretion of calcium in the urine)
- •Severe renal impairment
- •Hypervitaminosis D (excess vitamin D in the body)
- •Hypersensitivity to any component of the formulation
Drug Interactions
Corticosteroids
Can reduce calcium absorption.
Thiazide diuretics
May increase the risk of hypercalcemia by reducing calcium excretion.
Bisphosphonates and Fluoride
Calcium can interfere with their absorption. Separate administration by at least 30 minutes to 2 hours.
Cardiac glycosides (e.g., Digoxin)
Hypercalcemia may potentiate the toxic effects of cardiac glycosides.
Tetracycline and Quinolone antibiotics
Calcium can interfere with the absorption of these antibiotics. Administer at least 2-3 hours apart.
Storage
Store in a cool, dry place below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include nausea, vomiting, constipation, abdominal pain, thirst, polyuria, muscle weakness, and fatigue. Severe cases can lead to hypercalcemia, renal impairment, and cardiac arrhythmias. Management involves discontinuing the supplement, hydrating the patient, and monitoring serum calcium levels. In severe hypercalcemia, loop diuretics and corticosteroids may be used.
Pregnancy & Lactation
G-Calbo DX is generally considered safe and often recommended during pregnancy and lactation to meet increased calcium and vitamin D requirements. However, it should be used under medical supervision, and excessive doses should be avoided.
