Description
Overview of the medicine
Genac Plus is a combination medicine containing Paracetamol and Caffeine, used for rapid relief of various types of pain including headache, migraine, toothache, muscle pain, and fever. Caffeine enhances the pain-relieving effects of Paracetamol.
Uses & Indications
Dosage
Adults
1-2 tablets every 4-6 hours as needed, maximum 8 tablets in 24 hours.
Elderly
Same as adult dose, but caution for underlying conditions.
Renal_impairment
Dose adjustment may be necessary in severe renal impairment; consult a physician.
How to Take
Take orally with water. Can be taken with or without food. Do not exceed the recommended dose.
Mechanism of Action
Paracetamol works by inhibiting prostaglandin synthesis in the central nervous system, leading to analgesic and antipyretic effects. Caffeine acts as a CNS stimulant, vasoconstrictor, and enhances the analgesic effect of Paracetamol, possibly by increasing its absorption or direct action on pain pathways.

Onset
Pain relief usually within 15-30 minutes.
Excretion
Mainly via renal excretion as metabolites. Small amounts of unchanged Paracetamol are also excreted. Caffeine metabolites are excreted in urine.
Half life
Paracetamol: 1-4 hours (typically 2-3 hours). Caffeine: 3-7 hours.
Absorption
Both Paracetamol and Caffeine are rapidly and well absorbed from the gastrointestinal tract. Peak plasma concentrations of Paracetamol are reached within 30-60 minutes, and Caffeine within 30-120 minutes.
Metabolism
Paracetamol is primarily metabolized in the liver via glucuronidation and sulfation, with a small amount oxidized by cytochrome P450. Caffeine is extensively metabolized in the liver by CYP1A2.

Alcohol

Symptoms of overdose include nausea, vomiting, abdominal pain, sweating, drowsiness, and liver damage. Seek immediate medical attention. Treatment involves gastric lavage, activated charcoal, and N-acetylcysteine for Paracetamol overdose.
Pregnancy & Lactation
Paracetamol is generally considered safe in pregnancy at recommended doses. Caffeine intake should be moderate. Consult a doctor before use during pregnancy and lactation.
