Description
Overview of the medicine
Glipatab 10 mg Tablet contains Teneligliptin, an oral anti-diabetic drug used to improve glycemic control in adults with type 2 diabetes mellitus.
Uses & Indications
Dosage
Adults
Recommended dose is 10 mg once daily, taken orally. May be increased to 20 mg once daily if needed and tolerated.
Elderly
No dose adjustment required based solely on age.
Renal_impairment
No dose adjustment needed for mild to moderate renal impairment. For severe renal impairment or end-stage renal disease (ESRD), consider 5 mg once daily, but clinical data is limited.
How to Take
Take orally once daily, with or without food. It should be swallowed whole with water.
Mechanism of Action
Teneligliptin is a dipeptidyl peptidase-4 (DPP-4) inhibitor. It prevents the breakdown of incretin hormones (GLP-1 and GIP), thereby increasing their levels. This leads to glucose-dependent enhancement of insulin secretion and suppression of glucagon secretion, resulting in improved glycemic control.
Pharmacokinetics
Onset
Within hours of administration, sustained effect over 24 hours.
Excretion
Mainly excreted via kidneys (approx. 50%) and feces (approx. 50%).
Half life
Approximately 24 hours (effective half-life for DPP-4 inhibition).
Absorption
Rapidly absorbed after oral administration, peak plasma concentration reached within 1-2 hours.
Metabolism
Primarily metabolized by CYP3A4 and flavin-containing monooxygenase 3 (FMO3).
Side Effects
Contraindications
- Hypersensitivity to Teneligliptin or any component of the tablet
- Type 1 diabetes mellitus
- Diabetic ketoacidosis
Drug Interactions
Insulin or Sulfonylureas
Increased risk of hypoglycemia, may require dose adjustment of insulin/sulfonylureas.
CYP3A4/FMO3 inhibitors (e.g., Ketoconazole, Ritonavir)
May increase Teneligliptin plasma concentrations.
Storage
Store in a cool and dry place, below 30°C. Protect from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose are generally consistent with the known side effects. Management is symptomatic and supportive, including gastric lavage and observation.
Pregnancy & Lactation
Pregnancy Category B. Limited data in pregnant women. Use only if potential benefit justifies the potential risk to the fetus. It is not known whether Teneligliptin is excreted in human milk. Exercise caution when administered to a nursing woman.
