Description
Overview of the medicine
Glucodip 25 mg Tablet is an oral antidiabetic medicine used to treat type 2 diabetes mellitus. It belongs to a class of drugs called SGLT2 inhibitors. It can also be used to reduce the risk of cardiovascular death, hospitalization for heart failure, and progression of chronic kidney disease in specific patient populations.
Uses & Indications
Dosage
Adults
The recommended starting dose is 25 mg orally once daily in the morning, with or without food. The dose may be adjusted based on efficacy and tolerability.
Elderly
No dose adjustment is required based on age. However, greater sensitivity in some older individuals cannot be ruled out.
Renal_impairment
No dose adjustment is needed for mild to moderate renal impairment (eGFR ≥30 mL/min/1.73m²). Not recommended for severe renal impairment (eGFR <30 mL/min/1.73m²), end-stage renal disease (ESRD), or patients on dialysis for glycemic control. For cardiovascular and renal indications, may be initiated if eGFR ≥20 mL/min/1.73m².
How to Take
Take Glucodip 25 mg Tablet orally once daily in the morning, with or without food. Swallow the tablet whole with water. Do not crush, cut, or chew it.
Mechanism of Action
Glucodip works by blocking the sodium-glucose co-transporter 2 (SGLT2) protein in the kidneys. This action reduces the reabsorption of glucose from the kidney tubules back into the blood, leading to increased excretion of glucose in the urine. This helps to lower blood glucose levels.
Pharmacokinetics
Onset
Glucose-lowering effects typically observed within a few hours of administration.
Excretion
Approximately 75% of the dose is excreted via urine (primarily as metabolites) and about 25% via feces.
Half life
Approximately 10-15 hours, allowing for once-daily dosing.
Absorption
Rapidly absorbed after oral administration, reaching peak plasma concentrations (Cmax) within 1-2 hours. High oral bioavailability.
Metabolism
Extensively metabolized, primarily via glucuronidation by uridine diphosphate-glucuronosyltransferases (UGTs), predominantly UGT1A9.
Side Effects
Contraindications
- Hypersensitivity to the active substance or to any of the excipients.
- Severe renal impairment (eGFR <30 mL/min/1.73m²), end-stage renal disease (ESRD), or patients on dialysis for glycemic control.
- History of diabetic ketoacidosis.
Drug Interactions
Diuretics
Co-administration with diuretics may increase the risk of dehydration and hypotension. Close monitoring of volume status and blood pressure is recommended.
Insulin and Insulin Secretagogues (e.g., sulfonylureas)
Co-administration may increase the risk of hypoglycemia. A lower dose of insulin or sulfonylurea may be required.
Storage
Store in a cool and dry place, below 30°C. Protect from moisture and direct sunlight. Keep out of reach of children.
Overdose
In the event of an overdose, contact your doctor or emergency services immediately. Symptomatic and supportive treatment should be initiated as appropriate.
Pregnancy & Lactation
Pregnancy Category C. Glucodip is not recommended during pregnancy, especially during the second and third trimesters, due to potential adverse effects on fetal kidney development. It is also not recommended during lactation as it is unknown if it is excreted in human milk. Consult your doctor.

Clinical Trials
Extensive clinical trials, including cardiovascular outcome trials (CVOTs), have demonstrated the efficacy and safety of SGLT2 inhibitors in glycemic control, cardiovascular risk reduction, and renal protection in patients with type 2 diabetes, heart failure, and chronic kidney disease.
Lab Monitoring
- Renal function (eGFR) should be assessed before initiating treatment and monitored periodically thereafter.
- Blood glucose levels and HbA1c should be monitored regularly.
- Lipid profile may be monitored.
Doctor Notes
- Counsel patients on proper hydration and the importance of recognizing symptoms of genitourinary infections (e.g., candidiasis, UTI) and diabetic ketoacidosis (DKA).
- Monitor renal function (eGFR) at baseline and periodically thereafter, especially in elderly patients or those with pre-existing renal impairment.
- Advise patients about potential postural hypotension, particularly when co-administered with diuretics or antihypertensive agents.
Patient Guidelines
- Maintain adequate hydration to minimize the risk of volume depletion.
- Be aware of symptoms of genitourinary infections (e.g., pain during urination, increased frequency, unusual discharge) and diabetic ketoacidosis (e.g., nausea, vomiting, abdominal pain, rapid breathing, unusual tiredness).
- Inform your doctor or dentist about all medications you are taking, including over-the-counter drugs and herbal supplements.
Missed Dose Advice
If you miss a dose, take it as soon as you remember. However, if it is almost time for your next dose, skip the missed dose and resume your regular dosing schedule. Do not double the dose to catch up.
Driving Precautions
Glucodip may cause dizziness or lightheadedness in some patients, especially when starting treatment or if used with other blood pressure-lowering medications. Exercise caution when driving or operating machinery until you know how this medicine affects you.
Lifestyle Advice
- Adopt a healthy diet plan recommended by your doctor or dietitian.
- Engage in regular physical activity as advised by your healthcare provider.
- Maintain a healthy weight.
