Description
Overview of the medicine
Gluconor 2 mg Tablet contains Glimepiride, an oral antidiabetic drug in the sulfonylurea class. It is used to lower blood glucose levels in patients with type 2 diabetes mellitus when diet and exercise alone are not sufficient.
Uses & Indications
Dosage
Adults
Initially 1-2 mg once daily, administered with breakfast or the first main meal. Dosage can be increased gradually in increments of 1-2 mg at 1-2 week intervals based on blood glucose response, up to a maximum of 8 mg once daily.
Elderly
Initial dose of 1 mg once daily, titrated carefully.
Renal_impairment
Initial dose of 1 mg once daily. For severe renal impairment (CrCl < 22 mL/min), initial dose of 1 mg and careful titration is recommended.
How to Take
Take orally once daily with breakfast or the first main meal of the day. Swallow the tablet whole with water. Do not crush or chew.
Mechanism of Action
Glimepiride primarily lowers blood glucose by stimulating the release of insulin from pancreatic beta cells. It also increases the sensitivity of peripheral tissues to insulin.

Extensively metabolized in the liver by CYP2C9 to two main metabolites, a cyclohexylhydroxy derivative and a carboxyl derivative.

Contraindications
- Hypersensitivity to glimepiride or other sulfonylureas
- Diabetic ketoacidosis (with or without coma)
- Type 1 diabetes mellitus
- Severe renal or hepatic impairment (without dose adjustment)
Drug Interactions
NSAIDs, salicylates, sulfonamides, chloramphenicol, coumarins, probenecid, monoamine oxidase inhibitors, beta-blockers, alcohol
May potentiate the hypoglycemic effect of glimepiride.
Thiazides, corticosteroids, thyroid products, estrogens, oral contraceptives, phenytoin, nicotinic acid, sympathomimetics, calcium channel blockers
May reduce the hypoglycemic effect of glimepiride, leading to hyperglycemia.

Store in a cool, dry place, below 30°C. Protect from light and moisture. Keep out of reach of children.
