Jefenac 50 mg Tablet contains Aceclofenac, a non-steroidal anti-inflammatory drug (NSAID) used to relieve pain and inflammation associated with musculoskeletal disorders such as osteoarthritis, rheumatoid arthritis, and ankylosing spondylitis.
Uses & Indications
Dosage
Adults
The recommended dose is 100 mg twice daily. For Jefenac 50 mg, this translates to one 50 mg tablet twice daily or as directed by the physician.
Elderly
No dosage adjustment is generally required, but caution should be exercised due to increased risk of adverse effects in the elderly. The lowest effective dose should be used.
Renal_impairment
In mild to moderate renal impairment, dose adjustment may not be necessary. However, initiation with a lower dose and careful monitoring are advised. Avoid in severe renal impairment.
How to Take
Jefenac 50 mg Tablet should be swallowed whole with a glass of water, preferably with or after food, to minimize gastrointestinal discomfort. Do not chew or crush the tablet.
Mechanism of Action
Aceclofenac exerts its anti-inflammatory and analgesic effects primarily through the inhibition of prostaglandin synthesis. It inhibits both cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2) enzymes, leading to a reduction in the production of inflammatory mediators.

Onset
Onset of action is typically within 1-2 hours.
Excretion
Approximately two-thirds of the administered dose is excreted in the urine, mainly as hydroxylated metabolites, and one-third in the feces.
Half life
Plasma elimination half-life is approximately 4-4.3 hours.
Absorption
Rapidly and completely absorbed after oral administration. Peak plasma concentrations are reached in approximately 1.25 to 3 hours.
Metabolism
Extensively metabolized in the liver, primarily by CYP2C9, to 4'-hydroxyaceclofenac and other metabolites.
Side Effects
Contraindications
- •Hypersensitivity to aceclofenac, aspirin, or other NSAIDs
- •Patients with active gastrointestinal bleeding or peptic ulceration
- •Patients with severe heart failure, renal failure, or hepatic failure
- •Third trimester of pregnancy
- •History of bronchospasm, urticaria, or acute rhinitis precipitated by NSAIDs
Drug Interactions
SSRIs
Increased risk of gastrointestinal bleeding.
Lithium
Increased plasma lithium levels, leading to toxicity.
Methotrexate
Increased methotrexate toxicity.
Corticosteroids
Increased risk of gastrointestinal ulceration or bleeding.
Warfarin and other anticoagulants
Increased risk of bleeding.
Diuretics and antihypertensives (e.g., ACE inhibitors)
Reduced diuretic and antihypertensive effects.
Storage
Store below 30°C in a dry place, protected from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include nausea, vomiting, epigastric pain, gastrointestinal bleeding, headache, drowsiness. Management involves gastric lavage, activated charcoal, and symptomatic and supportive treatment. There is no specific antidote.
Pregnancy & Lactation
Avoid use during the third trimester of pregnancy due to potential for premature closure of the fetal ductus arteriosus. Use with caution in the first and second trimesters, and during lactation, only if the potential benefits outweigh the risks. Consult a doctor.
