Description
Overview of the medicine
Klavin 500 mg Tablet is a combination antibiotic containing Amoxicillin and Clavulanic Acid. It is used to treat a wide range of bacterial infections by killing the bacteria and preventing them from becoming resistant to the antibiotic.
Uses & Indications
Dosage
Adults
Typically 500 mg/125 mg tablet (Amoxicillin/Clavulanic Acid) every 8 hours, or 875 mg/125 mg every 12 hours, depending on the severity and type of infection. Max dose of Amoxicillin 6g/day, Clavulanic acid 600mg/day.
Elderly
Similar to adults, but dose adjustment may be necessary in case of impaired renal function.
Renal_impairment
Dosage should be reduced based on creatinine clearance. For CrCl 10-30 mL/min, 500 mg/125 mg every 12 hours. For CrCl <10 mL/min, 500 mg/125 mg every 24 hours. Hemodialysis patients: 500 mg/125 mg every 24 hours, plus an additional dose during and at the end of dialysis.
How to Take
Take orally at the start of a meal to minimize potential gastrointestinal intolerance. Do not crush or chew the tablet; swallow whole.
Mechanism of Action
Amoxicillin works by inhibiting bacterial cell wall synthesis, leading to bacterial cell death. Clavulanic Acid is a beta-lactamase inhibitor that protects Amoxicillin from degradation by beta-lactamase enzymes produced by some bacteria, thereby extending Amoxicillin's spectrum of activity.

Antibacterial effect typically observed within 1-2 hours.
Excretion
Primarily excreted unchanged by the kidneys (amoxicillin) and as metabolites by the kidneys (clavulanic acid).
Half life
Amoxicillin: approximately 1-1.5 hours. Clavulanic Acid: approximately 1 hour.
Absorption
Both amoxicillin and clavulanic acid are well absorbed from the gastrointestinal tract after oral administration. Peak plasma concentrations are reached in approximately 1 hour.
Metabolism
Amoxicillin is partially metabolized; clavulanic acid is extensively metabolized.
Side Effects
Contraindications
- •Hypersensitivity to penicillin or any component of the formulation
- •History of severe hypersensitivity reaction to any other beta-lactam antibiotic
- •History of jaundice/hepatic dysfunction associated with amoxicillin/clavulanate
Drug Interactions
Probenecid
Decreases renal tubular secretion of amoxicillin, increasing amoxicillin blood levels.
Allopurinol
Increased incidence of rash when co-administered with amoxicillin.
Methotrexate
May increase methotrexate toxicity by reducing its renal excretion.
Warfarin and other oral anticoagulants
May prolong prothrombin time (INR); monitor closely.
Storage
Store below 30°C in a dry place. Protect from light and moisture. Keep out of reach of children.
Overdose
Overdose symptoms are mainly gastrointestinal (e.g., nausea, vomiting, diarrhea) and disturbances of fluid and electrolyte balance. Crystalluria, leading to renal failure, has been observed in some cases. Management is supportive, and hemodialysis can be used to remove amoxicillin and clavulanic acid from circulation.
Pregnancy & Lactation
Pregnancy Category B. Studies in animals have not shown harm to the fetus, but there are no adequate and well-controlled studies in pregnant women. Use only if clearly needed. Excreted in breast milk; caution advised for breastfeeding mothers.
