Description
Overview of the medicine
Ktx 10 mg Tablet contains Ketorolac Tromethamine, a potent Non-steroidal Anti-inflammatory Drug (NSAID) primarily used for the short-term management of moderately severe acute pain that requires analgesia at the opioid level, usually in a post-operative setting. It provides effective relief from pain, inflammation, and fever.
Uses & Indications
Dosage
Adults
The recommended adult oral dose is 10 mg every 4-6 hours as needed, not to exceed 40 mg per day. Treatment duration should not exceed 5 days.
Elderly
A reduced dosage may be necessary, typically 10 mg every 6-8 hours, not to exceed 30 mg per day. Close monitoring is advised.
Renal_impairment
Dosage reduction is required. For moderate renal impairment, reduce the dose to 5 mg every 6-8 hours or avoid use in severe impairment.
How to Take
Take the tablet orally with a glass of water, preferably with food or milk to minimize gastrointestinal upset. Do not crush or chew the tablet.
Mechanism of Action
Ketorolac Tromethamine inhibits prostaglandin synthesis by blocking cyclooxygenase (COX-1 and COX-2) enzymes. This reduction in prostaglandin levels leads to analgesic, anti-inflammatory, and antipyretic effects.
Pharmacokinetics
Onset
Approximately 30 minutes for analgesic effect.
Excretion
Mainly renal (approximately 92% of the dose), with a small portion excreted in feces.
Half life
Approximately 4-6 hours (increases with renal impairment).
Absorption
Rapidly and completely absorbed orally. Peak plasma concentrations are achieved in approximately 30-60 minutes.
Metabolism
Primarily hepatic, mainly by glucuronidation.
Side Effects

- •Patients undergoing or who have recently undergone coronary artery bypass graft (CABG) surgery
- •Late pregnancy (third trimester) and labor
Drug Interactions
SSRIs
Increased risk of gastrointestinal bleeding.
Lithium
Increased plasma lithium levels, leading to toxicity.
Diuretics
Reduced diuretic effect and potential for increased risk of renal impairment.
Cyclosporine
Increased risk of nephrotoxicity.
Methotrexate
Increased methotrexate levels, leading to toxicity.
ACE Inhibitors/ARBs
Reduced antihypertensive effect and increased risk of renal impairment.
Anticoagulants (e.g., Warfarin)
Increased risk of bleeding.
Antiplatelet agents (e.g., Aspirin)
Increased risk of bleeding.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose may include nausea, vomiting, epigastric pain, lethargy, drowsiness, and gastrointestinal bleeding. Rarely, hypertension, acute renal failure, respiratory depression, and coma may occur. Management is primarily symptomatic and supportive; there is no specific antidote. Gastric decontamination (e.g., activated charcoal) may be considered if ingestion is recent.
Pregnancy & Lactation
Pregnancy Category C (first and second trimesters), Category D (third trimester). Avoid in the third trimester due to potential adverse effects on the fetus and inhibition of uterine contractions. Use with caution during lactation; small amounts may be excreted in breast milk.
