Description
Overview of the medicine
Lanozole 30 mg Capsule contains Lansoprazole, a proton pump inhibitor, used to reduce stomach acid production. It is prescribed for conditions like heartburn, acid reflux, stomach ulcers, and gastroesophageal reflux disease (GERD).
Uses & Indications
Dosage
Adults
GERD: 15-30 mg once daily for 4-8 weeks. Duodenal Ulcer: 15 mg once daily for 4 weeks. Gastric Ulcer: 30 mg once daily for 8 weeks. H. pylori eradication: 30 mg twice daily for 10-14 days (with antibiotics). Zollinger-Ellison Syndrome: Initial dose of 60 mg once daily, adjusted as needed.
Elderly
No specific dose adjustment is generally required, but caution is advised in patients with severe hepatic impairment.
Renal_impairment
No dose adjustment is necessary for patients with renal impairment.
How to Take
Take Lanozole capsule orally, preferably in the morning before breakfast. Swallow the capsule whole with water. Do not crush, chew, or open the capsule.
Mechanism of Action
Lansoprazole belongs to a class of drugs called proton pump inhibitors (PPIs). It works by irreversibly blocking the H+/K+-ATPase enzyme system (the proton pump) in the gastric parietal cells, thereby reducing gastric acid secretion.
Pharmacokinetics
Onset
Acid inhibition begins within 1 hour, maximum effect within 2-4 hours.
Excretion
Approximately two-thirds of the dose is excreted in the feces, and one-third in the urine as metabolites.
Half life
Plasma elimination half-life is 1.3-2.2 hours, but its acid-suppressing effect lasts much longer due to irreversible binding to the proton pump.
Absorption
Rapidly absorbed after oral administration, peak plasma concentrations occur within 1.5-2.2 hours. Bioavailability is about 80-90%. Food can delay absorption but does not significantly affect bioavailability.
Metabolism
Extensively metabolized in the liver by the cytochrome P450 enzyme system, primarily by CYP2C19 and CYP3A4. Metabolites are inactive.
Side Effects
Contraindications
- •Hypersensitivity to Lansoprazole or any component of the formulation
- •Concomitant use with nelfinavir (an antiretroviral drug)
Drug Interactions
Warfarin
Increased risk of bleeding due to inhibition of CYP2C19.
Clopidogrel
Reduced effectiveness of clopidogrel; consider alternative antiplatelet therapy.
Methotrexate
Increased plasma levels of methotrexate, especially with high doses.
Nelfinavir/Atazanavir
Reduced absorption and efficacy of these antiretroviral drugs.
Iron Salts/Ketoconazole
Reduced absorption due to altered gastric pH.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
There is no specific antidote for Lansoprazole overdose. Treatment should be symptomatic and supportive. Gastric lavage and activated charcoal may be considered if ingestion is recent.
Pregnancy & Lactation
Pregnancy Category B. Studies in animals have not shown a risk, but there are no adequate and well-controlled studies in pregnant women. Use only if clearly needed. It is unknown if Lansoprazole is excreted in human milk; use with caution in breastfeeding mothers.
