Overview of the medicine
Lasgrain 100 mg Tablet is a medication used for the acute treatment of migraine headaches with or without aura. It belongs to a class of drugs called triptans, which work by narrowing blood vessels in the brain and reducing substances that can trigger migraine symptoms.
Uses & Indications
Dosage
Adults
The usual recommended dose is 100 mg taken orally at the first sign of a migraine headache. If the migraine recurs or symptoms worsen after initial relief, a second 100 mg dose may be taken after at least 2 hours. Do not exceed a total dose of 200 mg within 24 hours.
Elderly
Use with caution in elderly patients (over 65 years) due to increased risk of cardiovascular events; consider a lower starting dose.
Renal_impairment
No dose adjustment is generally required for mild to moderate renal impairment. Use with caution in severe renal impairment.
How to Take
Take the tablet whole with water. It can be taken with or without food. Do not crush, chew, or split the tablet.
Mechanism of Action
Lasgrain acts as a selective agonist for 5-hydroxytryptamine1B and 5-hydroxytryptamine1D (5-HT1B/1D) receptors on cranial blood vessels and nerve endings. This action leads to vasoconstriction of intracranial blood vessels, which are dilated during a migraine attack, and inhibits the release of pro-inflammatory neuropeptides from trigeminal nerve endings, thereby aborting a migraine attack.

Onset
Onset of action typically occurs within 30-60 minutes.
Excretion
Excreted mainly via the kidneys, with a small portion eliminated in feces.
Half life
Elimination half-life is approximately 2-3 hours.
Absorption
Rapidly absorbed after oral administration, peak plasma concentrations reached within 1-2 hours.
Metabolism
Primarily metabolized in the liver, largely by monoamine oxidase (MAO) A.
Side Effects
Contraindications
- •History of ischemic heart disease (angina pectoris, myocardial infarction).
- •Uncontrolled hypertension.
- •History of stroke or transient ischemic attack (TIA).
- •Peripheral vascular disease.
- •Severe hepatic impairment.
- •Concomitant use with ergotamine-containing or ergot-type medications.
- •Concomitant use with other 5-HT1 agonists (triptans).
- •Hypersensitivity to the active substance or any excipients.
Drug Interactions
Cimetidine
Increases plasma concentration of Lasgrain.
Propranolol
May increase Lasgrain AUC.
SSRIs/SNRIs
Potential for serotonin syndrome; monitor patients.
MAO Inhibitors
Increased plasma levels of Lasgrain; contraindicated.
Ergotamine and Ergot-type drugs
Increased risk of vasospasm; contraindicated within 24 hours.
Other 5-HT1 agonists (triptans)
Increased risk of vasospasm; contraindicated within 24 hours.
Storage
Store below 30°C in a dry place, away from light and moisture. Keep out of reach of children.
Overdose
In case of overdose, symptoms may include severe dizziness, drowsiness, vomiting, and cardiovascular events. Treatment should be symptomatic and supportive, including monitoring of ECG and blood pressure. There is no specific antidote.
Pregnancy & Lactation
Pregnancy Category C. Use during pregnancy only if the potential benefit justifies the potential risk to the fetus. It is unknown whether Lasgrain is excreted in human milk; use with caution in breastfeeding mothers.
