Overview of the medicine
Laxpru 1 mg Tablet contains prucalopride, a medication used to treat chronic idiopathic constipation in adults. It works by increasing bowel motility and improving bowel function.
Uses & Indications
Dosage
Adults
The recommended dose is 1 mg once daily. If needed, and after assessment of efficacy and tolerability, the dose may be increased to 2 mg once daily. Treatment beyond 6 months should be re-evaluated.
Elderly
Initially 1 mg once daily. Adjust dosage based on individual response and tolerability. Caution advised for patients >75 years old.
Renal_impairment
For severe renal impairment (creatinine clearance <30 mL/min), the dose is 1 mg every other day. No dose adjustment is needed for mild to moderate impairment.
How to Take
Take orally with or without food, preferably at the same time each day.
Mechanism of Action
Prucalopride is a selective, high-affinity serotonin (5-HT4) receptor agonist. It stimulates the 5-HT4 receptors in the gastrointestinal tract, leading to increased colon motility, accelerating colonic transit, and promoting bowel movements.

Onset
Within 24-48 hours for the first spontaneous bowel movement.
Excretion
Mainly via urine (approximately 60-65% unchanged drug) and feces (approximately 20% unchanged drug).
Half life
Approximately 24-30 hours.
Absorption
Rapidly absorbed after oral administration. Absolute bioavailability is >90%. Peak plasma concentrations are reached within 2-3 hours.
Metabolism
Limited metabolism; primarily excreted unchanged. Metabolites account for less than 10% of total excretion.
Side Effects
Contraindications
- •Hypersensitivity to prucalopride or any excipients
- •Intestinal perforation or obstruction
- •Inflammatory bowel disease (Crohn's disease, ulcerative colitis)
- •Toxic megacolon/megarectum
Drug Interactions
Erythromycin
No clinically relevant interaction.
Ketoconazole
May slightly increase prucalopride plasma concentrations (minor effect, not clinically significant).
Oral contraceptives
No impact on the efficacy of oral contraceptives.
Storage
Store below 30°C in a dry place, protected from light and moisture. Keep out of reach of children.
Overdose
Symptoms may include headache, nausea, and diarrhea. In case of overdose, treatment is symptomatic and supportive. Gastric lavage may be considered in severe cases.
Pregnancy & Lactation
Prucalopride is not recommended during pregnancy. Use only if the potential benefit justifies the potential risk to the fetus. It is excreted in breast milk, so caution is advised during breastfeeding.
