Description
Overview of the medicine
Limpet 1 mg Tablet is an anxiolytic medication containing Zabaplon, indicated for the short-term treatment of generalized anxiety disorder and insomnia. It works by enhancing the effects of a natural chemical in the body (GABA) that calms brain activity.
Uses & Indications
Dosage
Adults
For GAD: 1 mg once daily, preferably at bedtime. May be adjusted to 0.5-2 mg/day based on response and tolerability. For insomnia: 1 mg at bedtime.
Elderly
Initial dose of 0.5 mg once daily, adjusted cautiously based on response. Elderly patients may be more sensitive to the effects.
Renal_impairment
Dose adjustment may be necessary in severe renal impairment. Start with a lower dose and monitor for adverse effects.
How to Take
Take Limpet 1 mg Tablet orally with a glass of water, with or without food. Do not crush, chew, or break the tablet.
Mechanism of Action
Zabaplon acts as a positive allosteric modulator of the GABA-A receptor. It binds to a specific site on the receptor, distinct from the GABA binding site, enhancing the inhibitory effects of GABA. This leads to increased chloride ion influx into neurons, resulting in hyperpolarization and reduced neuronal excitability.
Pharmacokinetics
Onset
Anxiolytic effects typically observed within 30-60 minutes.
Excretion
Excreted predominantly via the kidneys as metabolites.
Half life
Approximately 10-20 hours.
Absorption
Rapidly absorbed after oral administration, with peak plasma concentrations reached within 1-2 hours.
Metabolism
Primarily metabolized in the liver by cytochrome P450 enzymes, mainly CYP3A4.
Side Effects
Contraindications
- Hypersensitivity to Zabaplon or any components of the tablet
- Severe respiratory insufficiency
- Sleep apnea syndrome
- Acute narrow-angle glaucoma
- Myasthenia gravis
Drug Interactions
Opioids
Increased risk of severe sedation, respiratory depression, coma, and death.
Alcohol and CNS Depressants
Increased risk of sedation, respiratory depression, and hypotension.
CYP3A4 Inducers (e.g., rifampicin, carbamazepine)
May decrease plasma concentrations of Zabaplon, potentially reducing efficacy.
CYP3A4 Inhibitors (e.g., ketoconazole, erythromycin)
May increase plasma concentrations of Zabaplon, leading to enhanced effects.
Storage
Store at room temperature (below 30°C), protected from light and moisture. Keep out of reach of children.
Overdose
Symptoms of overdose include somnolence, confusion, diminished reflexes, and coma. Management involves supportive care, maintaining a patent airway, and monitoring vital signs. Flumazenil may be considered in severe cases, but caution is advised.
Pregnancy & Lactation
Pregnancy Category D. Zabaplon should be avoided during pregnancy, especially during the first trimester and near term, due to potential risk of congenital malformations and neonatal withdrawal symptoms. It is excreted in breast milk and should not be used during lactation.
