Uses & Indications
Dosage
Adults
For conjunctivitis: 1-2 drops into the affected eye(s) every 2-4 hours for the first two days, then 1-2 drops four times daily. For keratitis: 1-2 drops into the affected eye(s) every 30 minutes for the first two days, then 1-2 drops hourly for 4-6 days, then 1-2 drops four times daily.

Renal_impairment
No specific dosage adjustment required as systemic absorption is minimal.
How to Take
Wash hands before use. Tilt head back, pull down lower eyelid, and apply drops into the conjunctival sac. Close eye gently for a few minutes. Avoid touching the dropper tip to any surface.
Mechanism of Action
Ofloxacin inhibits bacterial DNA gyrase and topoisomerase IV, essential enzymes for bacterial DNA replication, transcription, repair, and recombination, leading to bacterial cell death.
Pharmacokinetics
Onset
Rapid; typically within minutes of administration.
Excretion
Primarily renal excretion for systemically absorbed drug.
Half life
Not well-defined for topical ophthalmic use due to minimal systemic absorption, but ranges from 4-7 hours systemically.
Absorption
Minimal systemic absorption after ocular administration. Achieves high concentrations in tear film and ocular tissues.
Metabolism
Limited hepatic metabolism; primarily excreted unchanged.
Side Effects
Contraindications
- Hypersensitivity to Ofloxacin or any other fluoroquinolone
- Hypersensitivity to any excipient of the eye drop
Drug Interactions
No significant systemic drug interactions expected due to minimal ocular absorption.
Topical corticosteroids might mask signs of infection and should be used with caution.
Storage
Store below 25°C. Do not freeze. Protect from light. Keep out of reach of children.
Overdose
Ocular overdose is unlikely to cause systemic toxicity due to limited absorption. Flush eye with lukewarm water if excessive amount is applied. Contact a healthcare professional if irritation persists.
Pregnancy & Lactation
Use during pregnancy only if the potential benefit outweighs the potential risk to the fetus. Caution is advised during lactation as Ofloxacin is excreted in breast milk after systemic administration, though minimal with topical eye drops.
